Expression and significance of tripartite motif protein 27 in tongue squamous cell carcinoma

目的探讨三结构域蛋白27(TRIM27)在舌鳞癌组织中的表达,以及下调该基因对细胞增殖、迁移和侵袭力的影响。方法选取2014-02~2017-02在武汉大学人民医院口腔科行手术治疗的舌鳞癌患者82例,免疫组化法检测舌鳞癌组织及癌旁组织中TRIM27蛋白表达。培养舌鳞癌SCC-9细胞并分为TRIM27干扰组、对照序列组和对照组,实时荧光定量PCR技术检测各组细胞中TRIM27基因表达,MTT法检测细胞增殖能力,Transwell法检测细胞迁移和侵袭能力。结果舌鳞癌组织中TRIM27蛋白阳性表达率为74.39%,癌旁组织中则为34.15%,两者间差异有统计学意义(χ2=40.104,P=0.000)。舌鳞癌组织中TRIM27蛋白阳性表达率在不同分化程度(中低分化与高分化)和是否有淋巴结转移中差异有统计学意义(P<0.05)。与对照序列组和对照组比较,TRIM27干扰组细胞中TRIM27 mRNA表达量降低(P<0.05),24 h,48 h,72 h和96 h时细胞吸光度值减低(P<0.05),且迁移细胞数及侵袭细胞数降低(P<0.05)。结论TRIM27蛋白在舌鳞癌组织中呈高表达,特异性下调舌鳞癌SCC-9细胞中TRIM27基因表达,可有效抑制细胞增殖、迁移和侵袭。

Objective To investigate the expression and significance of tripartite motif protein 27(TRIM27)in tongue squamous cell carcinoma,and to explore the effect of down-regulation of TRIM27 expression on cell proliferation,migration and invasion.Methods Eighty-two patients with tongue squamous cell carcinoma who underwent surgical treatment from February 2014 to February 2017 were selected in Department of Stomatology,Wuhan University People’s Hospital.Immunohistochemistry was used to detect the expression of TRIM27 protein in tongue squamous cell carcinoma and adjacent tissues.The tongue squamous cell carcinoma SCC-9 cells were cultured and divided into TRIM27 interference group,control sequence group and control group.Real-time fluorescence quantitative PCR was used to detect the expression of TRIM27 gene in the cells in each group.MTT assay was used to detect the cell proliferation.Transwell assay was used to detect the cell migration and cell invasion.Results The positive expression rate of TRIM27 protein was 74.39%in tongue squamous cell carcinoma and 34.15%in adjacent tissues(χ2=40.104,P=0.000).The positive expression rate of TRIM27 protein in tongue squamous cell carcinoma was significantly different between patients with middle-low differentiation and high differentiation,and between lymph node metastasis and non-metastasis patients(P<0.05).Compared with control sequence group and control group,the expression of TRIM27 mRNA was decreased in TRIM27 interference group(P<0.05),the A values at 24 h,48 h,72 h and 96 h were decreased(P<0.05),and the number of migrated cells and the number of invasive cells were also decreased(P<0.05).Conclusion TRIM27 protein is highly expressed in tongue squamous cell carcinoma tissues.Down-regulation of the TRIM27 expression in tongue squamous cell carcinoma SCC-9 cells could effectively inhibit cell proliferation,migration and invasion.
