强筋祛湿液药效学研究

Study on Pharmacodynamics of Qiangjin Qushi liquid

目的:研究院内制剂强筋祛湿液药效与安全性,以期为保证其临床应用安全与阐明药效提供科学依据。方法:最大给药量法考察强筋祛湿液家兔皮肤涂抹急性毒性;建立二甲苯致小鼠急性耳肿胀、角叉菜胶致大鼠足跖肿胀模型与佐剂性关节炎大鼠模型,观察强筋祛湿液对实验动物炎症反应的抑制作用;采用热板法观察强筋祛湿液对小鼠热板疼痛反应的影响。结果:急性毒性试验结果表明强筋祛湿液1日内最大给药量给药未见由药物引起的毒性反应;药效学表明强筋祛湿液对二甲苯所致小鼠耳肿胀、角叉菜胶致大鼠足跖肿胀与佐剂致大鼠关节炎的炎症反应具有显著的抑制作用(P<0.05),显著下调佐剂性关节炎大鼠血清中IL-6和TNF-α的水平(P<0.05),对热板所致的非特异性疼痛具有一定抑制作用(P<0.05)。结论:强筋祛湿液安全性较好,具有明显的抗炎和镇痛作用,可减轻滑膜增生、脂肪组织和滑膜上皮细胞增生、减少多种炎细胞浸润等多种病理学改变,且该作用与药物调节血清中IL-6和TNF-α水平有关。

Objective To provide a scientific basis for ensuring the safety of clinical application and elucidating the efficacy of Qiangjin Qushi liquid.Methods Study on acute toxicity of skin smear on rabbits with Qiangjin Qushi liquid by maximum dose method;The acute ear swelling in mice induced by xylene,paw swelling in rats induced by carrageenan and adjuvant arthritis in rats were established to observe the inhibitory effect of Qiangjin Qushi liquid on inflammatory reaction in experimental animals;and the effect of Qiangjin Qushi liquid on hot plate pain response in mice was observed by hot plate method.Results The acute toxicity test showed that the maximum dose of Qiangjin Qushi liquid in one day had no toxic reaction caused by drugs.Pharmacodynamics showed that Qiangjin Qushi liquid had significant inhibitory effects on xylene-induced ear swelling in mice,carrageenan-induced paw swelling in rats and adjuvant-induced arthritis in rats(P<0.05),and significantly decreased the serum level of adjuvant-induced arthritis in rats.The levels of IL-6 and TNF-alpha(P<0.05)inhibited the non-specific pain induced by hot plate(P<0.05).Conclusion Qiangjin Qushi liquid is safe and has good anti-inflammatory and analgesic effects.It can alleviate many pathological changes such as synovial hyperplasia,adipose tissue and synovial epithelial cell proliferation,and reduce a variety of inflammatory cell infiltration.This effect is related to the drug regulation of serum levels of IL-6 and TNF-α.
