摘要 目的:检测组氨酸三聚体核苷结合蛋白1(histidine triad nucleotide binding protein 1,Hint1)基因在小鼠早期妊娠过程中子宫内膜中的表达规律;探究Hint1基因在小鼠子宫内膜基质细胞蜕膜化过程的作用。方法:应用免疫组化、原位杂交、Western blot和RT-qPCR检测Hint1在早期妊娠、假孕和人工诱导蜕膜化模型小鼠子宫中的表达;siRNA敲低原代分离的小鼠子宫内膜基质细胞Hint1的表达。LC-MS分析Hint1对基质细胞蜕膜化过程中脂质组学的影响。结果:在妊娠的第1至第8天Hint1表达逐渐升高,且在胚胎植入部位的表达明显高于非植入点;体内人工诱导蜕膜化小鼠子宫内膜Hint1的表达明显高于未发生蜕膜化的对照子宫;利用siRNA干扰小鼠子宫内膜基质细胞中Hint1的表达可显著影响其蜕膜化过程及脂质代谢。结论:Hint1在基质细胞蜕膜化过程中表达升高,并可能与蜕膜化过程中基质细胞的脂质代谢相关。 Objective:To investigate the expression of histidine triad nucleotide binding protein 1(Hint1)gene in the endometrium during early pregnancy in mice and its role in decidualization of endometrial stromal cells. Methods:Immunohistochemistry,in situ hybridization,Western blot,and RT-qPCR were used to measure the expression of Hint1 in the uterus in mouse models of early pregnancy,pseudopregnancy,and artificially-induced decidualization,and siRNA was used to knockdown the expression of Hint1 in primary endometrial stromal cells. LC-MS was used to analyze the effect of Hint1 on lipidomic profile in stromal cells during decidualization. Results:The expression of Hint1 gradually increased from day 1 to day 8 of pregnancy,and the expression at implantation sites was significantly higher than that at non-implantation sites. The mice with artificially induced decidualization had significantly higher expression of Hint1 in the endometrium than the control mice without decidualization. The siRNA interference with Hint1 expression significantly affected decidualization and lipid metabolism in endometrial stromal cells. Conclusion :There is an increase in the expression of Hint1 in endometrial stromal cells during decidualization,which may be associated with lipid metabolism in endometrial stromal cells during decidualization.
