蛤蟆油对卵清蛋白诱发豚鼠哮喘的治疗作用及其机制研究 被引量:6

Effect of Oviductus Ranae on Ovalbumin Induced Asthma in Guinea Pigs and Its Mechanism

摘要 目的:探讨哈蟆油对哮喘模型豚鼠抗哮喘作用,并初步研究其作用机制。方法:将豚鼠随机分为空白组、模型组、地塞米松1 mg/kg组、蛤蟆油100 mg/kg、200 mg/kg、400 mg/kg组,并建立卵清蛋白特异性致敏诱导的豚鼠哮喘模型,分别灌胃给予各组相应药物或生理盐水,连续2周。酶联免疫法检测模型动物血清中炎性因子IL-4、IL-5、IL-13、Ig E的浓度;采用流式细胞术检测豚鼠肺泡灌洗液中嗜酸性粒细胞(EOS)的凋亡情况;血细胞分析仪检测血细胞计数;同时制备肺组织常规病理切片,观察模型动物肺组织病理学变化。结果:与空白组相比,模型组动物全血中EOS、NEU%、WBC细胞计数以及血清中IL-4、IL-5、IL-13、Ig E含量明显升高,蛤蟆油各组上述指标与模型组比较明显降低,其中200 mg/kg、400 mg/kg组动物血清中IL-4、IL-5、IL-13、Ig E含量较模型组显著下降。EOS凋亡比例显著增加,病理切片显示其炎性损伤程度较模型组有显著的改善。结论:蛤蟆油能够有效地降低模型动物血中IL-4、IL-5、IL-13、Ig E的含量,促使肺组织和气道壁嗜酸性粒细胞凋亡,恢复血液中EOS、NEU%、WBC的正常水平,改善肺组织病理损伤,对哮喘有一定的治疗作用。 Objective: To investigate the effect of Oviductus Ranae on asthma in guinea pigs and its mechanism. Methods: Guinea pigs were randomly divided into the control group,the model group,1 mg/kg dexamethasone group,Oviductus Ranae groups (400 mg/kg,200 mg/kg,100 mg/kg). Ovalbumin specific sensitization induced guinea pig asthma model was established,corresponding drugs and saline water were administered for 2 weeks. The serum concentrations of inflammatory cytokines IL-4,IL-5,IL-13 and IgE in the model animals were detected by ELISA. The apoptosis of eosinophil (EOS) cells in guinea pig alveolar lavage was detected by flow cytometry. The blood cell count was observed by hemacytometer. Routine pathological sections of lung tissues were prepared to observe the pathological changes. Results: Compared with the control group,EOS,NEU%,WBC cell count,levels of IL-4,IL-5,IL-13 and IgE of whole blood in the model group were increased. Compared with the model group,the above indexes in Oviductus Ranae group were reduced. Among them,the contents of IL-4,IL-5,IL-13 and IgE in 400 mg/kg and 200 mg/kg groups were significantly decreased. The results of EOS apoptosis were significantly increased.Pathological sections showed that the inflammatory injury degree in the drugs group was significantly improved. Conclusion: Oviductus Ranae can effectively reduce the contents of IL-4,IL-5,IL-13 and IgE in animal blood,it can also promote the apoptosis of eosinophils in lung tissues and airway wall,restore the normal level of EOS,NEU% and WBC in blood,improve the pathological injury of lung tissues,and has a certain therapeutic effect on asthma.
