内异方调控NF-КB p65改善大鼠子宫内膜异位症的机制研究 被引量:5

Improving Effect of Neiyi Recipe on Endometriosis via Regulating NF-κBp65 in Rats

摘要 目的:探讨内异方改善大鼠子宫内膜异位症(Endometriosis,EM)的作用机制。方法:构建大鼠EM模型,随机分成假手术组、模型组、内异方24. 8 g/kg、49. 6 g/kg、99. 2 g/kg组、孕三烯酮0. 25 mg/kg组,灌胃21天后剖解观测内异灶,ELISA法检测腹腔冲洗液中TNF-α、IL-1β、IL-2、IL-6的水平,qP CR、Western-blot法检测NF-κB p65基因、蛋白表达和磷酸化,免疫组化法检测NF-κBp65入核情况。结果:EM大鼠给予内异方治疗后,明显抑制內异灶的生长,改善盆腔粘连;显著降低了腹腔冲洗液TNF-α、IL-1β、IL-2、IL-6的含量;明显抑制NF-κB p65的基因、蛋白表达和磷酸化;显著抑制NF-κB p65核表达。结论:内异方可能通过抑制NF-κB p65的表达和活化,进而抑制炎症反应,抑制异位内膜的生长和黏附而改善EM。 Objective: To discuss the potential improving effect of Neiyi Recipe on endometriosis (EM) in rats and its mechanism. Methods: EM models of rats were established and randomly divided into the model group,Neiyi Recipe groups (24. 8 g/kg、49. 6 g/kg、99. 2 g/kg) and 0.25 mg/kg gestrinone group,some rats were set as the sham operation group. EM lesion was observed after 21 days of gavage administration.The levels of TNF-α,IL-1β,IL-2 and IL-6 were detected by ELISA,the expressions of NF-κBp65 and phosphorylation were detected by qP CR and western-blot,the expression of NF-κBp65 in the nucleus was detected by SP immunohistochemistry. Results: After treatment of Neiyi Recipe,the growth of EM lesion was significantly suppressed and pelvis adhesion was improved,the contents of TNF-α,IL-1β,IL-2 and IL-6 were significantly reduced,the expression and phosphorylation of NF-κBp65 were also significantly suppressed,and the expression of NF-κBp65 protein in the nucleus was significantly down-regulated. Conclusion: Neiyi Recipe can improve endometriosis through inhibiting the activity and expression of NF-κBp65,thus suppressing the inflammation reaction and inhibiting the growth of ectopic endometrium and adhesion.
