蒙药土茯苓-7汤对小鼠银屑病样皮损的干预作用及相关因子表达的影响 被引量:6

Effect of Mongolian Medicine Tufuling-7 Tang on Psoriasiform Skin Inflammation and Related Factors in Mice

摘要 目的:研究蒙药土茯苓-7汤对咪喹莫特诱导的银屑病样皮损的干预作用及皮损组织的增殖细胞核抗原(PCNA)、血管内皮生长因子(VEGF)、CD3^+、CD8^+等因子的表达的影响。方法:取BALB/C雄性小鼠,随机分成正常组、模型组、阳性对照组(消银颗粒1. 5 g/kg)、土茯苓-7汤0. 71、1. 42、2. 13 g/kg组。各组灌胃相应药物或生理盐水,每天一次,第6日药后采用药物诱导的方法激发小鼠皮肤出现银屑病样变化,再连续给药6日,处死小鼠,观察小鼠皮肤病损的(PASI)情况,光镜下观察皮损组织形态学变化,采用免疫组化法检测皮损组织的增殖细胞核抗原(PCNA)、血管内皮生长因子(VEGF)、CD3^+、CD8^+蛋白的表达。结果:模型组背部皮肤出现红斑、表皮肥厚、鳞屑成层等现象,PCNA、CD3^+、CD8^+、VEGF蛋白表达明显上调,蒙药土茯苓-7汤各剂量组银屑病样皮损减轻,PASI评分降低,角化不全现象得到改善,表皮厚度明显薄于模型组,其中2. 13 g/kg组改善最明显。与模型组比较,PCNA、CD3^+、CD8^+、VEGF蛋白表达结果显示蒙药土茯苓-7汤组均明显下调,具剂量依赖性。结论:蒙药土茯苓-7汤对咪喹模特诱导的小鼠银屑病样皮损具有明显的抑制作用。其作用机制为通过降低PCNA、VEGF、CD3^+、CD8^+蛋白的表达,抑制T淋巴细胞分化,缓解角质形成细胞的角化不全,角化过度,表皮异常增生等病理改变,抑制银屑病样皮损的发生。 Objective: To investigate the effect of Mongolian medicine Tufuling-7 tang on imiquimod-induced psoriasiform skin inflammation in Mice,and its effect on expressions of PCNA,VEGF,CD3^+、CD8 factors. Methods: BALB/C male mice were randomly divided into the control group,the model group,the positive control group (1. 5 g/kg Xiaoyin granule) and Tufuling 7 tang groups (0. 71、1. 42、2. 13 g/kg).Mice were administered with corresponding drugs or saline water each day,for 13 days. On 6 th day,psoriatic skin mice model was established by medicine. After administration,mice were sacrificed and psoriasis area and severity index PASI were observed. Histological changes of skin were observed by light microscope,the expressions of PCNA、VEGF、CD3^+、CD8^+were detected by immunohistochemistry. Results: In the model group,phenomena of erythema,skin thickening,scaly layering were observed on mice back skin. Expressions of PCNA、CD3^+、CD8^+、and VEGF were significantly up regulated. In Tufuling 7 tang groups,psoriasis-like lesions were alleviated、PASI score was decreased,parakeratosis was improved,skin thickening was reduced than that in the model group,while 2. 13 g/kg drug showed the best effect.Compared with the model group,expressions of PCNA、CD3^+、CD8^+、and VEGF were significantly down regulated in Tufuling 7 tang group,in dose dependant manner. Conclusion: Tufuling-7 tang has the inhibition on Psoriasis-like lesions of imiquimod-induced mice model,its mechanism may be down-regulating the expressions of PCNA、VEGF、CD3^+and CD8^+factors,inhibiting T lymphocyte differentiation,improving parakeratosis and epidermal hyperplasia,etc.
