藤茶源二氢杨梅素体外对破骨细胞的活性 被引量:1

The Activity of Ampelopsis Grossedentata Source Dihydromyricetin in Vitro on Osteoclasts

摘要 试验旨在研究不同浓度二氢杨梅素体外对破骨细胞活性的影响。用MTT方法检测不同浓度的二氢杨梅素对原代骨髓细胞生长存活率的影响。分离新西兰乳兔原代骨髓细胞,用1α, 25-(OH)2 Vit D3诱导获得破骨细胞,以雌二醇(10-8 mol/L)为阳性对照,根据骨吸收陷窝总面积、TRAP+细胞数及TRAP酶活性,分析藤茶源二氢杨梅素对破骨细胞活性的影响。结果表明,当二氢杨梅素浓度为10-5~10-8 mol/L时,骨髓细胞生长存活率接近100%。不同剂量二氢杨梅素对破骨细胞的活性均有抑制作用,其中以10-6和10-7 mol/L的剂量组对破骨细胞活性的抑制效果良好(p<0.01), 10-6 mol/L剂量组对破骨细胞活性的抑制效果最为显著(p<0.01)。藤茶源二氢杨梅素对破骨细胞活性有良好抑制作用。 It is to study the effects of different concentrations of dihydromyricetin on osteoclast activity in vitro. The effects of different concentrations of dihydromyricetin on the growth and survival rate of primary bone marrow cells were examined by MTT assay. The primary bone marrow cells of New Zealand milk rabbits were isolated, osteoclasts were induced by 1α, 25-(OH)2 VitD3 using estradiol(10-8 mol/L) as a positive control, and the effect of ampelopsis grossedentata source dihydromyricetin on osteoclast activity was analyzed based on the total area of bone resorption lacuna, TRAP+ cell number and TRAP enzyme activity. The results showed that the growth rate of bone marrow cells with dihydromyricetin concentration of 10-5 to 10-8 mol/L was close to 100%. The different doses of dihydromyricetin inhibited the activity of osteoclasts, and the inhibition of osteoclast activity was good in the dose groups of 10-6 mol/L and 10-7 mol/L(p<0.01). The 10-6 mol/L dose group had the most significant inhibitory effect on osteoclast activity(p<0.01). Dihydromyricetin from ampelopsis grossedentata has a good inhibitory effect on osteoclast activity.
