UPLC法同时测定人参中13种核苷类成分 被引量:5

Determination of 13 Nucleosides and Nucleobases in Different Parts of Different Ginseng by Ultra-high Performance Liquid Chromatography

摘要 试验建立了超高效液相色谱(Ultra Performance Liquid Chromatography, UPLC)法同时测定人参中13种核苷类成分含量的方法,研究了不同部位的鲜人参、生晒参、红参和林下参中核苷类成分含量的差异。采用超声辅助法提取人参样品中的核苷类成分,应用色谱柱Acquity UPLC HSS T3 (2.1 mm×100 mm, 1.8μm), 0.006%甲酸水溶液(A)-乙腈(B)梯度洗脱。鲜人参主根、芦头、须根中核苷类成分总含量分别为30.60, 33.44和27.29 mg/kg;生晒参3个部位核苷类成分总含量分别为83.62, 168.78和59.21 mg/kg;红参3个部位核苷类成分总含量分别为57.76, 121.98和2.62mg/kg;林下参3个部位核苷类成分总含量分别为213.21, 278.00和177.27 mg/kg。不同人参核苷类成分有所差异,林下参含量最高,生晒参次之,鲜人参最低;同一种人参不同部位芦头中核苷类成分含量最高,须根含量最低。 It was to establish a method of simultaneous determination of the content of 13 nucleosides and nucleobases in ginseng by ultra-high performance liquid chromatography(UPLC). The differences of nucleosides and nucleobases in fresh ginseng, white ginseng, red ginseng and similar wild ginseng were studied. The nucleosides and nucleobases in ginseng samples were extracted by water with assistance of ultrasound. An Acquity UPLC H-Class T3 column(2.1 mm×100 mm, ID 1.8 μm) was employed. The mobile phase consisted of 0.01% formic acid in water(A) and acetonitrile(B). The total contents of nucleosides and nucleobases in main root, fibrous root and root refs of fresh ginseng were 30.60, 33.44, and 27.29 mg/kg. And those three parts of white ginseng were 83.62, 168.78, and 59.21 mg/kg, which of red ginseng were 57.76, 121.98, and 42.62 mg/kg, and which of similar wild ginseng were 213.21, 278.00, and 177.27 mg/kg, respectively. The contents of nucleosides and nucleobases of different ginseng are different. The total content of nucleosides and nucleobases in different ginseng could be ranked in the following order: similar wild ginseng>white ginseng>red ginseng>fresh ginseng, and the content of nucleosides and nucleobases in different parts of the same ginseng could be ranked in the following order: root refs>main root>fibrous root.
