5-羟色胺、c-Fos蛋白及慢性心理应激对少数民族地区人群不良情绪的影响

Effects of 5-hydroxytryptamine,c-Fos protein and chronic psychological stress on negative emotion in individuals from ethnic minority areas

目的探讨血清5-羟色胺、c-Fos蛋白及慢性心理应激对少数民族地区人群不良情绪的影响,并研究两种血清学指标在慢性心理应激与不良情绪之间的作用。方法采用随机整群抽样方法,抽取广西南丹县少数民族地区的366名居民为研究对象。应用生活事件量表(LES)评估心理应激程度、简化症状自评量表评估不良情绪(焦虑和抑郁),同时采用酶联免疫吸附法检测血清5-羟色胺和c-Fos蛋白含量。分析c-Fos蛋白水平、5-羟色胺水平与LES总分、抑郁得分、焦虑得分和LES总分与抑郁得分、焦虑得分的相关性,以及5-羟色胺水平和c-Fos蛋白水平在心理应激与不良情绪之间的中介效应。结果5-羟色胺水平与LES总分、抑郁因子得分、焦虑因子得分均呈负相关,c-Fos蛋白水平与焦虑因子得分和抑郁因子得分均呈正相关,LES总分与焦虑因子得分和抑郁因子得分均呈正相关(均P<0.05)。多元线性回归分析结果显示,5-羟色胺对抑郁和焦虑均具有负向预测作用,c-Fos蛋白与LES总分对抑郁和焦虑均具有正向预测作用(均P<0.05)。Bootstrap法检验结果显示,5-羟色胺在心理应激和不良情绪之间存在中介效应,但c-Fos蛋白不存在中介效应。结论血清5-羟色胺、c-Fos蛋白水平及慢性心理应激与少数民族地区人群抑郁焦虑情绪之间紧密关联,5-羟色胺在慢性心理应激与抑郁和焦虑情绪之间起中介效应。

Objective To explore the effects of serum 5-hydroxytryptamine,c-Fos protein and chronic psychological stress on negative emotion in individuals from ethnic minority areas,and to study the effects of the two serological indexes on the relationship between chronic psychological stress and negative emotion.Methods A total of 366 residents from ethnic minority areas in Nandan County,Guangxi,were sampled as subjects using random cluster sampling method.Life Event Scale(LES)was used for evaluating the degree of psychological stress and Simplified Symptom Self-Rating Scale for evaluating negative emotion(including anxiety and depression),and concurrently serum 5-hydroxytryptamine and c-Fos protein contents were detected by enzyme-linked immunosorbent assay.The correlation of c-Fos protein/5-hydroxytryptamine level with total LES score,depression score or anxiety score,as well as the correlation of total LES score with depression score or anxiety score were analyzed,and moreover,the mediating effect of 5-hydroxytryptamine level or c-Fos protein level on the relationship between psychological stress and negative emotion were analyzed.Results The level of 5-hydroxytryptamine negatively correlated with total LES score,depression factor score and anxiety factor score,the level of c-Fos protein positively correlated with anxiety factor score and depression factor score,and total LES score positively correlated with anxiety factor score and depression factor score(all P<0.05).Multivariate linear regression analysis results showed that 5-hydroxytryptamine had a negative predictive effect on depression and anxiety,c-Fos protein and total LES score all had positive predictive effects on depression and anxiety(all P<0.05).The results of Bootstrap test showed that 5-hydroxytryptamine exerted a mediating effect on the relationship between psychological stress and negative emotion,but c-Fos protein had no mediating effect.Conclusion Serum 5-hydroxytryptamine level,c-Fos protein level and chronic psychological stress are closely related to depression and anxiety mood in individuals from ethnic minority areas,and 5-hydroxytry ptamine exerts a mediating effect on the relationship between chronic psychological stress and depression/anxiety mood.
