摘要
目的检测B2M基因在免疫性血小板减少性紫癜中的表达情况,探讨B2M的表达与免疫性血小板减少性紫癜临床相关性。方法收集本医院49例免疫性血小板减少性紫癜患者及50例健康个体的静脉全血,分离出单个核细胞(PBMCs),提取总RNA,并反转录获得cDNA,利用实时荧光定量PCR技术检测B2M基因mRNA表达情况;提取PBMCs中的蛋白,利用Wesrtern Blot技术检测B2M蛋白水平的表达情况。结果B2M基因在免疫性血小板减少性紫癜患者和正常健康个体中表达存在差异,有统计学意义(P<0.05)。结论B2M基因异常表达可能与免疫性血小板减少性紫癜的发生发展密切相关,具有一定的临床意义。
Objective To detect the expression of B2M gene in immune thrombocytopenic purpura so as to explore the clinical correlation between the expression of B2M and immune thrombocytopenic purpura.Methods The venous whole blood were collected from 49 patients with immune thrombocytopenic purpura and 50 healthy individuals.Then mononuclear cells(PBMCs)were isolated from these samples,total RNA was extracted,and cDNA was obtained by reverse transcription.Real-time PCR was used to detect the mRNA expression of B2M gene.At the same time,the protein in PBMCs was extracted,and the expression of B2M protein was detected by Wesrtern Blot.Results There was statistical significance on the difference in the expression of B2M gene between patients with immune thrombocytopenic purpura and normal healthy individuals(P<0.05).Conclusion The abnormal expression of B2M gene may be closely related to the occurrence and development of immune thrombocytopenic purpura,which has certain clinical significance.
