五因素混水平正交实验优化鱼鳞胶原提取工艺 被引量:7

摘要 本研究采用五因素混水平(L8(4^1×2^4))正交实验法优化酸法提取鱼鳞胶原蛋白的工艺。在其他外界条件不变的情况下,考查温度、酸的种类、料液比和提取时间对鱼鳞胶原蛋白提取率和纯度的影响,利用正交表设计各个因素和水平的组合方式,根据设计好的实施因子计划表执行试验。保证无关因素的统一。计算机分析得到试验直观分析表、效应曲线图和方差分析表,通过分析直观分析表可以找到试验的最优组合,通过效应曲线图分析各因素对于胶原蛋白提纯影响的重要性的差别,得到的因素重要性次序为C>D>B>A>E,通过极差分析得到因素水平配比A4B1C2D2E1,即25℃、0.5 mol/L的醋酸,以1∶20(g/mL)的料液比提取72 h为最佳的提取路线。最后进行数学建模,利用SAS软件得到提取率与各因素的函数关系。 In this study,five factors mixed level(L8(4^1×2^4))orthogonal experiment was used to optimize the acid extraction process of fish scale collagen.The effects of temperature,kinds of acid,solid-liquid ratio and extraction time on the extraction yield and purity of collagen were investigated by using the orthogonal table in the situation in which the other ambient condition is sure not to be changed.According to the implementation factor schedule,the combination moles of factors with levels were designed by orthogonal table,on the premise of assuring the unification of unrelated factors.The result was analysed by three methods:visual analysis table,effective curve and analysis of variance.The optimal combination of factor level has been obtained through the visual analysis table.The individual degree of the influence on the purification of collagen and the importance order(C>D>B>A>E)was obtained from the effect curve.The prime proportioning of factors(A4B1C2D2E1)was obtained through range analysis.that is 25℃,0.5 mol/L acetic acid,1∶20 solid-liquid ratio,and 72 h extraction time.Finally,the mathematical model is built,and the functional relationship between extraction rate and each factoris obtained by SAS software.
