摘要
目的探讨祛风解痉方药对哮喘模型小鼠气道高反应性及气道炎症的影响。方法以卵清蛋白滴鼻的方法建立哮喘小鼠模型,以祛风解痉方药进行干预,比较祛风解痉方药对哮喘模型小鼠肺脏组织病理学及气道高反应的干预效果。结果与空白对照组相比,模型组的Smith评分显著升高(P<0.05)。与模型组相比,中药低剂量组的Smith评分无显著性差异(P>0.05);而中药中剂量组、中药高剂量组以及阳性药物组的Smith评分显著降低(P<0.05)。在同一乙酰胆碱浓度下,与正常对照组相比,模型组的气道阻力显著升高(P<0.05);与模型组相比,高剂量中药组和阳性对照组的气道阻力显著降低(P<0.05)。与模型组相比,中药低剂量组、中药中剂量组、中药高剂量组以及阳性对照组的Vγ1 mRNA表达量显著降低(P<0.05)、Vγ4 mRNA表达量显著增加(P<0.05)。结论祛风解痉方药可改善哮喘小鼠模型气道高反应性,这与其抑制气道炎症及降低γδT细胞的Vγ1 mRNA表达量、增加Vγ4 mRNA表达量相关。
Objective To explore the effects of dispelling pathogenic wind for resolving convulsion prescription on airway hyperresponsiveness and airway inflammation in asthmatic model mice.Methods The asthmatic mice model was established by ovalbumin nasal dropping.After intervention of dispelling pathogenic wind for resolving convulsion prescription,the intervention effects of dispelling pathogenic wind for resolving convulsion prescription on lung histopathology and airway hyperresponsiveness in asthmatic mice were compared.Results Compared with the blank control group,the Smith score of asthma model group increased significantly(P<0.05).Compared with the model group,the Smith score of the low-dose Chinese medicine group had no significant difference(P>0.05),while the Smith score of the middle-dose Chinese medicine group,the high-dose Chinese medicine group and the positive group decreased significantly(P<0.05).At the same concentration of acetylcholine,the airway resistance of the model group was significantly higher than that of the normal control group(P<0.05);compared with the model group,the airway resistance of the high-dose Chinese medicine group and the positive drug control group was significantly lower(P<0.05).Compared with asthma model group,the expression of Vγ1 mRNA were significantly decreased(P<0.05)and Vγ4 mRNA were significantly increased in low-dose Chinese medicine group,middle-dose Chinese medicine group,high-dose group and positive group(P<0.05).Conclusion Dispelling pathogenic wind for resolving convulsion prescription can improve airway hyperresponsiveness in asthmatic mice model,which is related to its inhibition of airway inflammation and decreasing the expression of Vγ1 mRNA and increasing the expression of Vγ4 mRNA inγδT cells.
