Six1和MTDH在人脑胶质瘤中的表达、临床病理特征及预后的相关性分析 被引量:1

摘要 目的探讨Six1和异黏蛋白(MTDH)在人脑胶质瘤中的表达、患者临床病理特征及预后的相关性。方法选取该院2012年12月至2016年12月收治的150例人脑胶质瘤患者作为研究组,同时选择60例非肿瘤脑组织作为对照组。采用免疫组化法检测脑组织中Six1和MTDH蛋白表达水平,采用χ2检验分析Six1和MTDH蛋白表达与脑胶质瘤患者临床病理特征的关系,并采用Cox比例风险分析脑胶质瘤患者预后影响因素。结果研究组Six1、MTDH蛋白阳性表达率[65.33%(98/150)、63.33%(95/150)]均明显高于对照组[33.33%(20/60)、28.33%(17/60),P<0.05]。Six1、MTDH蛋白表达与脑胶质瘤患者的分化程度差异有统计学意义(P<0.05);Spearman等级相关分析显示:脑胶质瘤患者中Six1与MTDH蛋白表达呈正相关(r=0.354,P<0.05);脑胶质瘤患者平均中位生存期48个月,其中Six1阳性表达和阴性表达患者中位生存期分别为21个月、44个月,2组比较差异有统计学意义(χ2=22.872,P<0.05);MTDH阳性表达和阴性表达患者中位生存期分别为19个月、45个月,2组比较差异有统计学意义(χ2=24.361,P<0.05);单因素分析显示:病理学分级越高、肿瘤低分化、Six1阳性、MTDH阳性患者其生存时间均明显缩短(P<0.05);多因素Cox回归模型分析显示:肿瘤低分化、Six1阳性、MTDH阳性均为脑胶质瘤患者预后的独立危险因素(P<0.05)。结论Six1与MTDH蛋白在脑胶质瘤患者组织中的表达水平升高,其表达水平随着人脑胶质瘤的恶性程度升高而升高,Six1表达阳性或MTDH表达阳性患者预后较差。 Objective To investigate the expression of Six1 and metadherin(MTDH)in human glioma and its correlation with clinicopathological features and prognosis.Methods A total of 150 cases of human glioma patients admitted to our hospital from December 2012 to December 2016 were selected as the study group,while 60 cases of non-tumorous brain tissue were selected as the control group.The expression of Six1 and MTDH in brain tissue was detected by immunohistochemical.The relationship between the expression of Six1 and MTDH and the clinicopathological characteristics of glioma patients were analyzed byχ2,the prognostic factors of glioma patients were analyzed by Cox proportional risk analysis.Results The positive expression rates of Six1 and MTDH protein in the study group[65.33%(98/150)、63.33%(95/150)]were significantly higher than those in the control group[33.33%(20/60),28.33%(17/60),P<0.05].The expression of Six1 and MTDH was significantly difference with the differentiation degree of glioma patients(P<0.05).Spearman grade correlation analysis showed that there was a positive correlation between the expression of Six1 and MTDH in glioma patients(r=0.354,P<0.05).The median survival time of glioma patients was 48 months,and the median survival time of patients with positive and negative expression of Six1 was 21 months and 44 months,respectively,the difference was significant(χ2=22.872,P<0.05).The median survival time of MTDH positive and negative expression patients was 19 months and 45 months,respectively,with significant difference between the two groups(χ2=24.361,P<0.05).Univariate analysis showed that the survival time of patients with higher pathological grade,lower differentiation of tumors,Six1 positive and MTDH positive was significantly shortened(P<0.05).Multivariate Cox regression analysis showed that low differentiation,Six1 positive and MTDH positive were independent risk factors for the prognosis of glioma patients(P<0.05).Conclusion The expression levels of Six1 and MTDH protein in glioma tissues increased with the malignant degree of human glioma.The prognosis of patients with positive expression of Six1 or MTDH was poor.
