参灵口服液的研制及其提高免疫力作用的研究 被引量:3

摘要 目的:采用正交试验筛选参灵口服液的最佳配方,并观察参灵口服液提高免疫力的作用。方法:以三七、西洋参、黄芪、甘草、枸杞、灵芝、马鹿茸为原料,分别超声制备总多糖和总皂苷,选用料液比(多糖、皂苷与水)、木糖醇、蜂蜜及柠檬酸4个因素,进行L9(3^4)正交设计,筛选参灵口服液配方。按照最佳配方制备参灵口服液,40只昆明种小鼠被随机分为空白对照组与参灵口服液高、中、低剂量组,小鼠灌胃10 d后处死,称取胸腺和脾脏的体质量,并进行ConA诱导小鼠脾淋巴细胞转化实验,计算ConA^+与ConA^-差值。结果:参灵口服液的最佳配方为料液比1∶20,木糖醇1.4 g,蜂蜜1.0 g,柠檬酸0.04 g。参灵口服液可显著增高小鼠胸腺指数和脾指数(P<0.05),提高ConA+与ConA-差值(P<0.05)。结论:参灵口服液色泽棕褐,澄清透明,酸甜适口,各理化指标符合相关国家标准。参灵口服液能促进ConA诱导小鼠脾淋巴细胞转化,具有良好的增强免疫力的作用。 Objective:To screen optimum formula of Shenling oral liquid by orthogonal test,and observe its improving immune function.Methods:First,total polysaccharide in ganoderma lucidum,cervus elaphus linnaeus,lycium barbarum,astragalus and licorice were extracted while total saponin in panax notoginseng and panax quinquefolium were extracted.Then optimum formula of Shenling oral liquids was studied through orthogonal design of L9(3^4),material-liquid ratio(polysaccharide,saponin and water),xylitol,honey and citric acid as four factors.Finally,Shenling oral liquids were prepared according to the optimum formula.40 mice were randomly divided into blank control group and high,medium and low dosage groups of Shenling oral liquids(0.4 mL/10 g,0.3 mL/10 g,0.2 mL/10 g),after 10 days administration mice were executed,thymus and spleen were weighed and splenic lymphocyte transformation experiment induced by ConA were conducted.Results:The optimum fomula was as follows:material-liquid ratio 1∶20,xylitol 1.4 g,honey 1.0 g,citric acid 0.04 g.Compared with the blank control group,index of thymus and spleen was increased and the difference between ConA+and ConA-was increased(P<0.05).Conclusion:Shenling oral liquid which was prepared according to the optimum formula was clear brown liquid,sweet and sour,no odor,and chemical indices were compounded with relevant national standards.Shenling oral liquids could improve splenic lymphocyte transformation induced by ConA,so possess a good role in enhancing immunity,had a good ability to improve immunity.
