新生鼠油对大鼠烫伤创面的治疗作用及可能机制研究

摘要 目的:探究新生鼠油对大鼠深Ⅱ度烫伤创面的治疗作用及作用机制。方法:120只SD大鼠随机分为新生鼠油组、京万红软膏组、烫伤模型组和正常对照组。对新生鼠油组、京万红软膏组、烫伤模型组大鼠背部造成深Ⅱ度烫伤,伤后立即给药,1次/d,连续15 d。在烫伤后1、3、5、7、10、15 d取材,测定药物对各组大鼠烫伤创面愈合率、组织含水量、羟脯氨酸(HYP)含量的影响;测定药物对烫伤大鼠血清肿瘤坏死因子α(TNF-α)、白细胞介素1(IL-1)水平的影响。结果:新生鼠油可不同程度地提高大鼠烫伤创面愈合率,降低烫伤创面组织含水量,升高烫伤创面HYP含量,降低烫伤大鼠血清TNF-α和IL-1水平。结论:新生鼠油对大鼠深Ⅱ度烫伤具有良好的保护作用,能明显减少创面组织含水量,促进烫伤创面愈合,其机制与增加创面组织HYP的合成与释放,降低烫伤大鼠血清中TNF-α、IL-1水平有关。 Objective:To study the therapeutic effect of Newborn Mice Oil on deep partial-thickness burn in rats,and to explore its mechanism.Methods:One hundred and twenty SD rats were randomly divided into Newborn Mice Oil group,Jingwanhong ointment(京万红软膏)group,scald model group and normal control group.The deep gradeⅡskin burn was made in Newborn Mice Oil group,Jingwanhong ointment group and Scald model group.The drug was adopted for wounds once everyday for 15 days.Samples were collected at 1st,3rd,5th,7th,10th,15th day after burn to determine the wound healing rate,the wound tissues water content,HYP,TNF-αand IL-1.Results:At different time points,Newborn Mice Oil could increase the wound healing rate and HYP content and decrease the tissues water content,TNF-αand IL-1.Conclusion:Newborn Mice Oil shows a notable protective effect on deep gradeⅡburn wounds in rats by obviously promoting the wound healing and decreasing the wound tissues water content.Its mechanisms may be related with the increase in HYP synthesis and release of wound tissues and the decrease in TNF-αand IL-1 in rat blood serum.
