摘要
1,3,4-三磷酸肌醇5/6激酶(ITPK)作为磷脂酰肌醇信号通路的一种重要的调控酶,在动物、植物、线虫的生物信号传导和生长发育中起到重要作用。为了进一步对毛果杨中ITPK家族基因进行系统分析,利用毛果杨的基因组数据库,通过生物信息学方法,对毛果杨ITPK基因家族的基因结构、染色体位置、蛋白结构域、细胞定位预测、系统进化、基因表达进行了分析。结果表明:毛果杨中的ITPK家族基因共有6个成员,分布于毛果杨的6条染色上。进化树分析显示毛果杨ITPK基因可分为3个亚家族;基于保守域分析显示,Pt ITPK1~PtITPK5均含有3个保守结构域,Pt ITPK6只含有2个保守结构域;毛果杨ITPK基因在各个器官或组织中均有表达。研究结果为进一步克隆该家族基因与验证ITPK家族基因与生物逆境胁迫相关提供科学依据。
As an important regulatory enzyme in the phosphatidylinositol signal pathway, 1,3,4-triphosphoinositol5/6 kinase(ITPK) plays an important role in the biological signal transduction and growth and development of animals, plants and nematodes. In order to analyze the ITPK family gene of Populus trichocarpa further systematically, the genome database of Populus trichocarpa used to analyze the gene structure, chromosome position,protein domain, subcellular localization, systematic evolution and gene expression of the ITPK gene family of Populus trichocarpa by bioinformatics methods. The results showed that there are six members totally in the ITPK family genes of the populus which distribute on six chromosomes in Populus trichocarpa. Evolutionary tree analysis shown that ITPK gene of the Populus trichocarpa can be divided into 3 subfamilies. According to the conservative domain analysis, PtITPK1~PtITPK5 contained three conservative structural domains, and PtlTPK6 contained two conservative domains. The ITPK gene of Populus trichocarpa is expressed in all organs and tissues. The results provide scientific basis, which is further cloning the family genes and verifying the relationship between ITPK family genes and biological stress.
