摘要
目的肿瘤的产生和发展是多项因素共同作用的结果,其中,免疫系统起着至关重要的作用。整合素相关蛋白(cluster of differentiation 47,CD47)是一种广泛表达于多种细胞表面的跨膜蛋白。CD47高表达于众多肿瘤细胞表面,其受体信号调节蛋白α(signal regulatory proteinα,SIRPα)主要表达在巨噬细胞表面,使癌细胞免于被巨噬细胞杀伤,造成免疫逃逸。另外,CD47-SIRPα通路也影响着异种器官移植的成功。解决不同种属生物间CD47与SIRPα一般不结合问题,可明显提高异种器官移植的成功率。本文从CD47与SIRPα的生理结构、抗肿瘤机制、相关药物的研发情况以及该通路在异种器官移植中的作用进行综述,并对该通路目前的研究进展进行介绍。
Objective The occurrence and development of tumors are the results of multiple factors,in which the immune system plays a vital role.Cluster of differentiation 47(CD47)is a transmembrane protein that widely expressed on the surface of a variety of cells,which is highly expressed on the surface of many tumor cells,while its receptor,signal regulatory proteinα(SIRPα),is mainly expressed on the surface of macrophages.Their combination can protects the cancer cells from being killed by macrophages,resulting in immune escape.In addition,the CD47-SIRPαpathway also affects the success of xenotransplantation.Solving the problem of non-combination of CD47 and SIRPαamong different species of organisms may greatly improve the success rate of xenotransplantation.This article reviews the physiological structure and anti-tumor mechanism of CD47 and SIRPα,the development of related drugs,and the role of this pathway in xenotransplantation,and by this way to introduce the current research progress of this pathway.
