抗菌活性肽六肽对糖尿病小鼠器官病理损伤影响的观察 被引量:1

Effect of antimicrobial peptide RRWWCR on pathological lesions in diabetic mice

摘要 目的观察抗菌活性肽六肽(RRWWCR)对糖尿病小鼠各器官病理损伤的影响。方法STZ腹腔注射建立糖尿病小鼠模型。成模后在小鼠背部脊柱上方不被自身舔舐和挠伤的部位形成机械性皮肤损伤,并随机分为伤口擦拭给药RRWWCR高浓度组(H,1000μg/ml)、中浓度组(M,200μg/ml)、低浓度组(L,40μg/ml)和不给药对照组(Con),每组各15只。间隔4周以同样方式再次构建皮肤创伤和擦拭给药,重复4次。每次给药前检测FBG。当Con组小鼠出现死亡时,每组随机处死小鼠5只,取肝、脾、肺、肾等组织制备石蜡切片及HE染色,观察组织病理改变。结果与Con组比较,M组FBG水平明显降低(P<0.01),具有减轻糖尿病小鼠肝、脾、肺、肾等组织器官损伤的作用。结论浓度为200μg/ml的RRWWCR对糖尿病小鼠器官病理损伤有减轻作用。 Objective To observe the effect of antimicrobial peptide(AMPs)(Arg Arg Trp Trp Cys Arg,RRWWCR)on pathological lesions in diabetic mice.Methods A diabetic mouse model was established by intraperitoneal injection of streptozotocin(STZ).Thereafter,mechanical skin injuries were created on the back where the mice will not be rubbed and scratched by themselves.The injured skin were randomized into 4 treatment groups:high RRWWCR concentration(H group,1000μg/ml),medium concentration(M group,200μg/ml),low concentration(L group,40μg/ml)and non-administered group(Con).15 mice in each group were treated with corresponding concentration of RRWWCR solution.Using the same way,skin damage and wiping administration were carried out after 4 weeks and were repeated 4 times.Before each administration,fasting blood glucose(FBG)from tail vein of the mice was detected.When the mice in Con group began to die,5 mice in each group were sacrificed at random.The liver,spleen,lung and kidney tissue were taken to observe histo-pathological changes using HE staining Results Compared with Con group,the FBG level in M group was significantly decreased(P<0.01),and the degree of damage to liver,spleen,lung and kidney tissue was smaller.Conclusion The AMP RRWWCR with a concentration of 200μg/ml can alleviate the pathological damage in diabetic mice.
