羧甲淀粉钠溶液对地塞米松诱导的免疫低下幼鼠的影响

Effect of carboxymethyl starch sodium solution on the immune suppression induced by dexamethasone in immature mice

摘要 目的考察羧甲基淀粉钠(carboxymethyl starch sodium,CMS)溶液对地塞米松诱导BALB/c幼鼠免疫低下模型的免疫调节作用。方法 10-12g幼鼠随机分为对照组、模型组、玉屏风组、CMS溶液低及中及高剂量组,每组10只,对照组与模型组纯净水灌胃,其他组每日分别给于玉屏风1.8g/kg,CMS溶液3.5g/kg、7.0g/kg和14.0g/kg,D8幼鼠腹腔注射地塞米松建立免疫低下模型,末次给药后24 h,称重,取血,解剖,取胸腺和脾脏计算脏器指数,检测血常规、自然杀伤(natural killer,NK)细胞杀伤活力、吞噬百分率、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)和白介素-6(interleukin-6,IL-6)水平。结果与正常组比较,模型组幼鼠体质量、胸腺和脾脏指数、白细胞计数和淋巴细胞百分比、NK细胞杀伤活性、吞噬百分率、TNF-a和IL-6均降低(P<0.05),血小板计数(PLT)和中性粒细胞百分比均有显著性升高(P<0.05);与模型组比较,CMS溶液高剂量组幼鼠体质量显著性升高(P<0.05),胸腺指数增高(P<0.05),PLT降低(P<0.05),高剂量组NK细胞杀伤活力升高(P<0.05),吞噬细胞活性和TNF-α分泌水平有一定提高,并且中、高剂量组与模型组比较有显著差异(P<0.05),CMS溶液高剂量组与玉屏风组相比各指标均无明显差异(P<0.05)。结论 CMS溶液灌胃给药可明显改善免疫低下幼鼠的体质量、胸腺指数、PLT、NK细胞杀伤活性和吞噬细胞活性,具有提高机体免疫力作用。 Objective To investigate the effect of carboxymethyl starch sodium(CMS) solution on immune regulation of dexamethasone-induced hypoimmunity in BALB/c mice. Methods Young mice weighing 10-12 g were randomly divided into control group, model group, Yupingfeng group, CMS solution low, medium and high dose groups, 10 mice in each group, the control group and the model group were given pure water by gavage. The other groups were respectively given Yupingfeng granule of 1.8 g/kg, CMS solutions of 3.5 g/kg, 7.0 g/kg and 14.0 g/kg each day. On day 8, mice were intraperitoneally injected with dexamethasone to establish a hypoimmune model. 24 h after the last dose, the body was weighed, blood was taken, the body was dissected, and the thymus and spleen were taken to calculate the organ index. Blood routine, natural killer(NK) cell killing activity, phagocytosis percentage, TNF-α and IL-6 levels were detected. Results Compared with the normal group, the body weight, thymus and spleen indexes, white blood cell and percentage of lymphocytes, NK cell killing activity, phagocytosis percentage, TNF-α and IL-6 level were all decreased in the model group(P<0.05), platelet count(PLT) and neutrophil percentage were significantly increased(P<0.05). Compared with the model group, the body weight of mice in the CMS solution high-dose group increased significantly(P<0.05), thymus index increased(P<0.05), and PLT decreased(P<0.05). NK cell killing activity increased in the high dose group(P<0.05), phagocyte activity and TNF-α secretion level increased to a certain extent, and there were significant differences between the medium and high dose groups and the model group(P<0.05). There were no significant differences in each index between the CMS solution high dose group and the Yupingfeng group. Conclusion Carboxymethyl starch sodium solution given by intragastric administration can significantly improve the body weight, thymus index, PLT, NK cell killing activity and phagocytic cell activity of immunocompromised young mice, and has the effects of improving the body immunity.
