摘要
目的:探讨雌激素受体β(ERβ)在膀胱癌中的表达及对膀胱癌细胞侵袭、迁移的影响。方法:收集我院2017年1至12月60例膀胱癌根治术患者的膀胱癌组织标本及相应的癌旁组织标本,采用免疫组织化学法测定膀胱癌及癌旁组织中ERβ表达。将T24细胞分为空白对照组(不转染)、阴性对照组(转染阴性对照siRNA)和si-ERβ组(转染ERβsiRNA)。采用Western blot法测定T24细胞ERβ、N-cadherin、Vimentin和基质金属蛋白酶9(MMP9)蛋白水平。采用划痕实验和Transwell小室测定T24细胞侵袭、迁移能力。结果:膀胱癌组织中ERβ阳性率明显高于癌旁组织(P<0.05)。膀胱癌细胞T24中ERβ相对表达量高于正常膀胱上皮细胞SV-HUC-1(P<0.05)。与空白对照组和阴性对照组比较,si-ERβ组T24细胞中ERβ相对表达量、侵袭和迁移能力降低(P<0.05),N-cadherin、Vimentin、MMP9蛋白水平降低(P<0.05),空白对照组和阴性对照组T24细胞中ERβ相对表达量、侵袭和迁移能力、N-cadherin、Vimentin、MMP9蛋白水平比较差异无统计学意义(P>0.05)。结论:膀胱癌组织中ERβ呈高表达,下调ERβ可抑制膀胱癌细胞的侵袭和迁移,其机制可能与沉默ERβ抑制上皮间质转化(EMT)有关。
Objective:To investigate the expression of estrogen receptor β(ERβ) in bladder cancer and its effect on invasion and migration of bladder cancer cells.Methods: The bladder cancer tissue specimens and the corresponding specimens of adjacent cancer tissues from the 60 cases of patients who underwent radical surgery for bladder cancer were collected from January 2017 to December 2017 in our hospital.The expression of ERβ in the bladder cancer and the adjacent tissues were determined by immunohistochemistry.The T24 cells were divided into blank control group(no transfection),negative control group(transfected with negative control siRNA) and si-ERβ group(transfected with ERβ siRNA).The levels of ERβ,N-cadherin,Vimentin and matrix metalloproteinase 9(MMP9) in T24 cells were determined by Western blot.Transwell chamber was used to measure the invasion and migration ability of T24 cells.Results: The positive rate of ERβ in bladder cancer tissues was significantly higher than that in adjacent tissues(P<0.05).The relative expression of ERβ in bladder cancer cell line T24 was higher than that in normal bladder epithelial cells SV-HUC-1(P<0.05).Compared with the blank control group and the negative control group,the relative expression of ERβ,the invasion and migration ability in T24 cells of the si-ERβ group were decreased(P<0.05),and the levels of N-cadherin,Vimentin and MMP9 protein expression of the si-ERβ group were decreased(P<0.05).There were no significant difference in the relative expression of ERβ,invasion and migration ability,the protein levels of N-cadherin,Vimentin and MMP9 between the blank control group and the negative control group(P>0.05).Conclusion: ERβ is highly expressed in the bladder cancer tissues.Down-regulation of ERβ can inhibit the invasion and migration of bladder cancer cells,and its mechanism may be related to the inhibition of epithelial-mesenchymal transition(EMT) by silencing ERβ.
