甘草次酸对人乳腺癌MDA-MB-231细胞的影响研究 被引量:12

摘要 目的研究甘草次酸对乳腺癌MDA-MB-231细胞的影响研究。方法 将MDA-MB-231细胞分为空白组、实验组和对照组,实验组以终浓度为20μmol·L^-1的甘草次酸处理,对照组以5μmol·L^-1的依托泊苷处理,空白组以等体积生理盐水处理。以MTT法检测乳腺癌MDA-MB-231细胞增殖,以Wound Healing法检测乳腺癌MDA-MB-231迁移能力,以蛋白免疫印迹(WB)法检测乳腺癌MDA-MB-231细胞磷酸化细胞外调节蛋白激酶(p-ERK)、基质金属蛋白酶-9(MMP-9)蛋白表达。结果 干预24 h后,空白组、实验组和对照组MDA-MB-231细胞活率分别为(99. 65±0. 21)%,(64. 64±6. 18)%,(60. 16±8. 42)%,干预48 h后分别为(98. 26±0. 32)%,(45. 12±4. 25)%,(36. 18±6. 47)%;干预48 h时,空白组、实验组和对照组实验组划痕愈合率分别为(64. 17±4. 67)%,(35. 48±5. 12)%,(32. 45±6. 14)%,细胞p-ERK1蛋白相对表达量分别为0. 85±0. 12,0. 52±0. 11,0. 36±0. 12,MMP-9蛋白相对表达量分别为0. 69±0. 06,0. 41±0. 12,0. 36±0. 10,差异均有统计学意义(均P <0. 05)。结论 甘草次酸可显著抑制乳腺癌MDA-MB-231细胞增殖和迁移,可能与抑制ERK1/2通路活化,下调MMP-9蛋白表达有关。 Objective To investigate the effect of glycyrrhetinic acid on breast cancer MDA-MB-231 cells.Methods MDA-MB-231 cells were divided into blank group,test group and control group.The test group was treated with glycyrrhetinic acid at the final concentration of 20μmol·L^-1,the control group was treated with etoposide at the concentration of 5μmol·L^-1,and the blank group was treated with normal saline at the same volume.MTT assay was used to detect the proliferation of breast cancer MDA-MB-231 cells,Wound Healing method was used to detect the migration ability of breast cancer MDA-MB-231 cells,and protein expression of phosphorylated extracellular regulatory protein kinase(p-ERK)and matrix metalloproteinase-9(MMP-9)in breast cancer MDA-MB-231 cells was detected by Western blot(WB).Results After 24 h of intervention,the viability of MDA-MB-231 cells in blank group,test group and control group were(99.65±0.21)%,(64.64±6.18)%,(60.16±8.42)%,respectively,and which were(98.26±0.32)%,(45.12±4.25)%,(36.18±6.47)%after 48 h of intervention.At 48 h of intervention,the healing rates of scratches in blank group,test group and control group were(64.17±4.67)%,(35.48±5.12)%,(32.45±6.14)%,the relative expression of p-ERK1 protein were 0.85±0.12,0.52±0.11,0.36±0.12,and the MMP-9 protein were 0.69±0.06,0.41±0.12,0.36±0.10,respectively.The difference were statistically significant(all P<0.05).Conclusion Glycyrrhetinic acid can significantly inhibit the proliferation and migration of breast cancer MDA-MB-231 cells,which may be related to the inhibition of ERK1/2 pathway activation and the down-regulation of MMP-9 protein expression.
