两种枸杞对干旱胁迫的生理响应及抗旱性评价 被引量:21

摘要 【目的】研究黑果枸杞和‘宁杞1号’在不同程度干旱胁迫下的生理响应,筛选抗旱评价指标,综合评价材料之间的抗旱能力差异.【方法】以2种枸杞组培幼苗为材料,采用盆栽控水法人工模拟轻度、中度和重度干旱胁迫,测定叶片的12个抗旱相关生理指标,结合隶属函数法与主成分分析筛选适合抗旱评价的指标,并进行抗旱能力综合评价.【结果】随着胁迫程度的加剧,供试材料的可溶性糖、可溶性蛋白、Pro、总黄酮和总酚含量均持续上升,POD和SOD活性均呈先上升后下降变化;O2·-含量、H2O2含量、MDA含量和APX活性均呈先下降后上升变化.主成分分析结果表明,O2·-含量、H2O2含量、可溶性糖含量、SOD活性和总酚含量5个指标的载荷量大,分别为-0.852 0、-0.856 6、0.962 7、0.905 6和0.922 5,可作为鉴定黑果枸杞和‘宁杞1号’抗旱性强弱的关键指标.【结论】通过隶属函数值结合主成分分析的抗旱性综合指数显示:黑果枸杞的抗旱综合指数为0.466,抗旱性高于‘宁杞1号’(0.446),两种枸杞均属于中度抗旱树种. 【Objective】 In order to study the physiological responses of Lycium ruthenicum and L.bararum cv. ‘NingqiⅠ’ under different degrees drought stress. The drought resistance evaluation indexes were selected and drought resistance ability differences between materials were comprehensively evaluated. 【Method】 Two kinds of wolfberry clones were used as material by using pot control water method to artificial simulation of mild,moderate and severe drought stress,12 related physiological indexes were measured,the subordinate function value method and principal component analysis were used to estimate drought resistance evaluation indexes and drought resistance comprehensive evaluation. 【Result】 The content of soluble sugar,soluble protein,proline,total flavonoids and total phenols of the two materials increased continuously with the increase of drought stress,and the POD and SOD activities increased first and then decreased with the increase of drought stress. The content of superoxide anion,hydrogen peroxide,malondialdehyde and APX activity decreased first and then increased with the deepening of drought stress. The principal component analysis showed that the loads of the five indexes such as O2 content,H2O2 content,soluble sugar content,SOD activity and total phenol content were-0.852 0,-0.856 6,0.962 7,0.905 6 and 0.922 5 respectively,which could be used as the key indexes to identify the drought resistance of L. ruthenicum and L. bararum cv. ‘NingqiⅠ’. 【Conclusion】 The drought resistance index of L. ruthenicum(0.466) was stronger than that of L. bararum cv. ‘NingqiⅠ’(0.446),and the two wolfberries belonged to moderate drought resistance tree species.
