育胞汤对卵巢功能不全大鼠卵巢VEGF、IFN-γ表达的影响

Effect of Yubao decoction on expressions of VEGF and IFN-γ in ovary of rats with premature ovarian insufficiency

目的:观察中药育胞汤对雷公藤多苷片诱导卵巢功能不全(POI)大鼠卵巢血管内皮生长因子(VEGF)、干扰素-γ(IFN-γ)表达的影响。方法:30只SD雌性大鼠,经雷公藤多苷片给药造模成功后随机分为3组,即中药育胞汤组、西药克罗米芬组和空白对照组。采用免疫组化法检测大鼠卵巢组织VEGF和IFN-γ的表达水平。结果:育胞汤组能提高POI大鼠卵巢内VEGF的表达,降低大鼠卵巢IFN-γ的表达,与空白对照组比较,差异有统计学意义(P﹤0.05)。结论:育胞汤可以促进卵巢功能不全的恢复,其作用机制可能为育胞汤可以促进卵巢血管形成,改善卵巢微循环,调节降低细胞因子水平,延缓卵泡闭锁及优势卵泡募集,促进卵泡发育,从而改善卵巢功能。

Objective:To observe the effect of Yubao decoction of Chinese medical herbs on the expressions of vascular endothelial growth factor(VEGF)and interferon-γ(IFN-γ)in ovary of rats with premature ovarian insufficiency(POI)induced by tripterygium wilfordii polyglycoside tablets,and to explore the mechanism of Yuebao decoction of Chinese medical herbs on POI.Methods:30 female SD rats were modeled successfully by tripterygium wilfordii polyglycoside tablets,then randomly divided into three groups,i.e.Yubao decoction group,clomiphene(western medicine)group and blank control group.The expressions of VEGF and IFN-γin ovarian tissue of rats were detected by immunohistochemistry.Results:Yubao decoction group could increase the expression of VEGF in ovary of POI rats,and reduce the expression of IFN-γin ovary of POI rats.Compared with the blank control group,the difference was statistically significant(P﹤0.05).Conclusion:Yubao decoction can promote the recovery of ovarian insufficiency.Its mechanism may be that Yuebao decoction can promote ovarian angiogenesis,improve ovarian microcirculation,regulate and reduce cytokines levels,delay follicular atresia and superior follicular recruitment,promote follicular development,thereby improve ovarian function.
