‘西拉’葡萄果实发育过程中糖代谢相关基因的表达分析 被引量:10

摘要 为了分析‘西拉’(Syrah)葡萄果实转色期至成熟期的发育过程中可溶性糖的积累与糖代谢相关基因表达量的关系,以山西临汾地区‘西拉’葡萄5个发育时期的果实为试材,测定葡萄果实发育过程中还原糖、总酸以及葡萄果皮酚类物质的质量分数,并利用实时荧光定量RT-PCR的方法,分析果实发育过程中蔗糖转运蛋白、单糖转运蛋白以及糖代谢相关酶(蔗糖合酶、蔗糖磷酸合酶和转化酶)等15个基因的表达量以及可溶性糖(葡萄糖和果糖)质量分数的变化趋势。结果表明:葡萄糖、果糖质量分数随着果实发育逐渐增加,己糖转运蛋白合成基因VvHT1表达量呈逐渐下降的趋势,在果实发育的后期下降较快;VvHT2和VvHT5表达量呈逐渐上升的趋势;VvHT3和VvMSA的表达量呈“降-升-降”的趋势,两者的表达量均在花后71 d最高。蔗糖运输蛋白(VvSUC11,VvSUC12,VvSUC27)合成基因的表达模式在发育过程中存在差异,VvSUC11和VvSUC12表达水平上调,VvSUC27呈下调的表达趋势。蔗糖转运蛋白、转化酶和单糖转运蛋白等的协同表达促进‘西拉’葡萄果实中可溶性糖的积累,在‘西拉’葡萄果实发育过程中发挥着重要的作用。 To analyze the relationship of soluble sugar accumulation with sugar-related gene expression during Syrah fruit development from veraison to the harvest,‘Syrah’fruit sampled at five developmental stage in Linfen,Shanxi province was used as the test material to determine the mass fraction of reducing sugar,total acid and phenolics.And real-time RT-PCR primers were designed to analyse both changes of the mRNA expression of fifteen genes including sucrose transporters,monosaccharide transporters and sugar metabolism-related enzymes(sucrose synthase,sucrose phosphate synthase,and invertase)and soluble sugars(glucose,fructose)content,respectively.The main results showed that the contents of glucose and fructose increased with the development of fruit.The mRNA expression level of hexose transporter VvHT1 decreased gradually,and a rapid decrease occurred at the late stage of fruit development;the mRNA expression level of VvHT2 and VvHT5 increased gradually;the mRNA expression level of VvHT3 and VvMSA showed a trend of“down-up-down”,and both mRNA expression levels were the highest at 71 d after flowering.The the sucrose transporters(VvSUC11,VvSUC12,VvSUC27)hadvariousexpression patterns during fruit development,the expression levels of VvSUC11 and VvSUC12 were up-regulated,and the expression of VvSUC27 was down-regulated.The synergistic expression of sucrose transporters,invertases and monosaccharide transporters promote the accumulation of soluble sugar in the fruit of‘Syrah’grapes,and they play an important role in regulation of‘Syrah’grape ripening.
