摘要
目的探究糖耐量正常人(Normal glucose tolerance,NGT)粪样微生物移植(Fecal microbiota transplantation,FMT)对胰岛素抵抗(Insulin resistance,IR)小鼠糖脂代谢的影响。方法根据粪样微生物移植纳入排除标准,筛选合适供体,收集粪样并按照干预方式处理粪样,-80℃保存。将造胰岛素抵抗模型成功小鼠按照体质量随机分为4组:高脂饮食组(High fat diet,HFD),高脂+供体粪样共生菌液组(High fat diet+Donor fecal symbiotic bacteria liquid,HFD+DFSBL),高脂+供体粪样菌悬液组(High fat diet+Donor fecal bacteria suspension,HFD+DFBS),高脂+供体粪样无菌滤液组(High fat diet+Donor fecal sterile filtrate,HFD+DFSF),尾尖采血检测各组小鼠的随机血糖、空腹血糖、血浆总胆固醇和甘油三酯水平,并进行口服葡萄糖耐量实验和胰岛素耐量实验。结果干预8周后,HFD+DFSBL组小鼠总胆固醇含量与HFD组相比,差异具有统计学意义(P<0.05);HFD+DFSBL组和HFD+DFBS组小鼠口服葡萄糖耐量实验曲线下面积与HFD组相比,差异有统计学意义(P<0.01)。结论糖耐量正常人粪样共生菌对胰岛素抵抗小鼠糖脂代谢的影响高于粪样中共生菌的代谢产物的影响。
Objective To investigate the effects of Normal glucose tolerance(NGT)fecal microbiota transplantation(FMT)on glycolipid metabolism in insulin resistance(IR)mice.Methods According to the excretion criteria of fecal microbial transplantation,selected appropriate donors and collected their fecal samples,treated fecal samples by means of intervention and stored the samples at-80℃.Based on body weight,the successful IR model mice were randomly divided into four groups:high fat diet(HFD)group,high fat diet+donor fecal symbiotic bacteria liquid(HFD+DFSBL)group,High fat diet+donor fecal bacteria suspension(HFD+DFBS)group and High fat diet+donor fecal sterile filtrate(HFD+DFSF)group.The blood glucose in every group was detected randomly.Fasting blood glucose,total plasma cholesterol and triglycerides levels and oral glucose tolerance test and insulin tolerance test were measured.Results After 8weeks of intervention,the total cholesterol in the HFD+DFSBL group was significantly different from that in the HFD group(P<0.05).The area under the curve of oral glucose tolerance test in the HFD+DFSBL group and the HFD+DFBS group were significantly different from that in the HFD group(P<0.01).Conclusion The effect of normal glucose tolerance fecal microbiota on glycolipid metabolism in the insulin resistant mice is higher than that of microbial metabolites on it.
