摘要
目的以2型糖尿病模型db/db小鼠为研究对象,探讨二甲双胍及粪菌移植技术对其肠微生物Faecalibacterium prausnitzii(F.prausnitzii)菌的影响。方法将db/m作为正常对照组,db/db小鼠分为db/db模型组、db/db+二甲双胍(db/db+Met)组和db/db+哈萨克族正常糖耐量人粪菌液组(db/db+kngt)组。采用16Sr DNA-PCR技术检测各组小鼠粪样中F.prausnitzii菌水平,分析F.prausnitzii菌水平与2型糖尿病相关表型的相关性。结果(1)与db/m正常对照组相比,db/db模型组粪样中F.prausnitzi菌水平显著降低,差异具有统计学意义(P<0.01);二甲双胍干预后可明显升高小鼠粪样中F.prausnitzi菌的水平,差异具有统计学意义(P<0.05);db/db+kngt组中F.prausnitzi水平有所升高,差异无统计学意义。(2)小鼠粪样中F.prausnitzii菌水平与小鼠FBG、TC、TG呈明显负相关(r=-0.812,r=-0.761,r=-0.707)。结论肠道F.prausnitzii菌水平与2型糖尿病的发生与改善密切相关。
Objective To investigate the effects of metformin and fecal microbiota transplantation technique on Faecalibacterium prausnitzii(F.prausnitzii)in type 2diabetes mellitus model of db/db mice.Methods db/m mice was as normal control group,and db/db mice were divided into db/db model group,db/db+metformin(db/db+met)group,db/db+Kazakh normal glucose tolerance human fecal liquid group(db/db+kngt)group.The level of F.prausnitzii in the feces of each group was detected by 16Sr DNA-PCR;the correlation between the level of F.prausnitzii and the phenotype associated with type 2diabetes was analyzed.Results(1)Compared with the db/m group,the F.prausnitzii level in the fecal samples of the db/db model group was significantly lower,and the difference was statistically significant(P<0.01);metformin could significantly increased the level of F.prausnitzii in mice feces after the intervention of it,and the difference was statistically significant(P<0.05);the level of F.prausnitzii in db/db+kngt group was increased,but the difference was not statistically significant.(2)Correlation analysis:The level of F.prausnitzii in mice feces was significantly negatively correlated with fasting blood glucose,total cholesterol and triglyceride in mice(r=-0.812,-0.761,and-0.707,all P<0.01).Conclusion The level of F.prausnitzii in the intestine is closely related to the occurrence and improvement of type 2diabetes.
