miR-126的细胞周期调控分析

摘要 目的 观察miR-126在肝癌细胞中的表达及其与细胞周期调控的关系,对miR-126在肝细胞肝癌的临床应用中的生物学功能进行初步探究.方法 选取45例肝癌患者作为本次实验的观察对象,分别利用荧光定量PCR及原位杂交法对所选取45例肝癌患者的癌细胞及其癌旁组织进行检测,观察miR-126的表达水平,并利用流式细胞仪对患者肝癌细胞进行细胞周期检测,分析miR-126的表达水平与细胞周期的关系.结果 45例肝癌患者的肝癌组织标本中的miR-126相对表达量为癌旁正常组织的(17.73±9.21)%,在癌旁正常组织中的阳性表达率为80.00%(36/45);38例高中分化肝癌组织中的miR-126相对表达量为癌旁正常组织的(19.88±8.81)%,而7例低分化肝癌患者的miR-126相对表达量为癌旁正常组织的(9.15±4.76)%;45例肝癌患者的肝癌组织标本中的miR-126在肝癌组织中的阳性表达率为31.11%(14/45);其中具有高中分化肝癌13例,低级分化肝癌1例;肝癌组织与癌旁正常组织细胞周期G1/G0期比例随着肝癌细胞中的S期的增加而降低(P<0.05);肝癌组织miR-126的阳性表达率与阴性表达率也具有显著差异,G1/G0期比例随着肝癌细胞中的S期的增加而降低(P<0.05).结论 miR-126在肝癌组织中表达水平较低且随着肿瘤分化进程的下降而降低,该改变方式对肝癌细胞的分裂与增殖起到促进作用. Objective To observe the expression of microRNA-126 in hepatocellular carcinoma cells and its relationship with cell cycle regulation,and to explore the biological function of microRNA-126 in clinical application of hepatocellular carcinoma.Methods Forty-five patients with hepatocellular carcinoma were selected as the subjects of this experiment.The expression of microRNA-126 was detected by fluorescence quantitative PCR and in situ hybridization in the cancer cells and adjacent tissues of 45 patients with hepatocellular carcinoma.The cell cycle of hepatocellular carcinoma cells was detected by flow cytometry,and the relationship between the expression of microRNA-126 and cell cycle was analyzed.Results The relative expression of microRNA-126 in 45 cases of hepatocellular carcinoma was(17.73±9.21)%,80.00%(36/45)in normal tissues adjacent to cancer,(19.88±8.81)%in 38 cases of moderately differentiated hepatocellular carcinoma,and(9.15±4.76)%in 7 cases of poorly differentiated hepatocellular carcinoma.The positive expression rate of microRNA-126 was 31.11%(14/45)in hepatocellular carcinoma tissues of 45 patients with hepatocellular carcinoma(HCC).Among them,13 cases were highly differentiated HCC and 1 case was low differentiated HCC.The ratio of G1/G0 phase of cell cycle between HCC tissues and adjacent normal tissues decreased with the increase of S phase in HCC cells(P<0.05).The positive and negative expression rates of 126 were also significantly different.The proportion of G1/G0 phase decreased with the increase of S phase in hepatocellular carcinoma cells(P<0.05).Conclusion The expression of microRNA-126 in hepatocellular carcinoma tissues is low and decreases with the decline of tumor differentiation,which promotes the division and proliferation of hepatocellular carcinoma cells.
