摘要 【目的】以乌龙茶提取物作为醋酸双氧铀的替代物用于真菌样品的电子染色,使其能达到与醋酸双氧铀一样的染色效果。【方法】利用酿酒酵母制备超薄切片样品,再在复染铅的情况下对超薄切片样品进行乌龙茶提取物以及醋酸双氧铀2种不同染料的染色,之后在透射电镜下观察比较其不同之处。【结果】乌龙茶提取物-铅复染法可得到与醋酸双氧铀-铅复染法极为相近反差效果的超微结构。【结论】乌龙茶提取物可作为既安全又方便易得的醋酸双氧铀替代物用于生物样品的超薄切片染色。 To check whether it is possible to use oolong tea extract(OTE) as a substitute for uranyl acetate(UA) in observation the ultrastructures of Saccharomyces cerevisiae.Two electron microscopic staining methods(UA and OTE)were compared for electron microscopic observation of ultrathin sections with post stained by lead citrate(Pb).The OTE+Pb staining method showed the results were similar to that of standard method with UA+Pb.This study demonstrated that OTE could potentially be used as a safer and more easily to get alternative to UA in electron microscopy staining for ultrathin sections of biological samples.
