摘要
目的解决血袋离心过程中下坠、折叠、血袋中红细胞挂壁现象,提高脐带血有核细胞回收率,确保脐带血有核细胞数量。方法2018年7月-9月,采用珍珠棉泡沫板作为支撑,进行三步离心分离浓缩脐带血有核细胞,计算有核细胞回收率及红细胞回收率,并与常规方法进行对比分析。结果采用设计方式放置采血袋进行离心后血袋表面平整,血袋未下坠折叠,细胞分层较为清晰。相较常规方法,单份制备时间则由(18.22±11.46)min降至(13.46±5.86)min,差异有统计学意义(P<0.05)。有核细胞回收率达到(84.27±5.96)%,差异有统计学意义(P<0.01)。筛选大于100 mL的大体积标本,平均TNC回收率达(83.06±5.71)%,RBC平均回收率为(23.68±8.06)%,质量显著提升(P<0.05),但CD34+细胞数无显著提高(P>0.05)。结论采用珍珠棉泡沫板支撑血袋,有效解决了离心过程中血袋下坠、折叠问题,减少了细胞损失,提高了制备效率及质量。
Objective To solve the phenomenon of falling,folding and red blood cell(RBC)remaining on the inner wall of the blood bags during centrifugation,improve the recovery rate of umbilical cord blood nucleated cells and ensure the number of nucleated cells in umbilical cord blood.Methods From July to September 2018,the pearl cotton EPE foamed plate was used as the support to centrifuge and concentrate the cord blood nucleated cells by three steps centrifugation.The recovery rate of nucleated cells and the recovery rate of red blood cells were calculated and compared with the traditional methods.Results After centrifugation,the surface of the blood bag was smooth,and the blood bag had not fallen down,and the cell layer was clear.Compared with the traditional method,the daily average preparation amount increased from(16.6±6.89)bags to(21.1±8.26)bags,while the preparation time decreased from(18.22±11.46)min to(13.46±5.86)min.The difference was statistically significant(P<0.05).The recovery rate of nucleated cells and RBCs were(84.27±5.96%)and(32.49±14.23%),respectively.The difference was statistically significant(P<0.01).Screening large volume samples larger than 100 ml,the average recovery rate of TNC and RBC were(83.06±5.71%)and(23.68±8.06%),respectively.The quality of the samples was significantly improved(P<0.05),but the number of CD34+cells was not significantly increased(P>0.05).Conclusion The use of pearl cotton foam board to support blood bags can effectively solve the problem of blood bags falling and folding during the centrifugal process,and greatly reduce the cell loss and improve the preparation efficiency and quality.
