摘要
目的按照L9(34)正交方案,最终以多糖含量作为长白山野生牛肝菌提取率指标,并检测其体外抗氧化活性.方法对长白山野生牛肝菌子实体多糖提取条件(液料比、提取时间、提取温度、提取次数)进行优化,检测其体外抗氧化活性.以长白山野生牛肝菌为原料,应用水杨酸法测定以蒸馏水为溶剂的水溶性维生素E和以蒸馏水为溶剂的长白山野生牛肝菌提取多糖清除羟基自由基的能力.结果 4个因素对长白山野生牛肝菌多糖提取率的影响表现为液料比(A,R=2.66)>提取时间(B,R=1.73)>提取温度(C,R=1.02)>提取次数(D,R=0.19),确定了长白山野生牛肝菌多糖提取的最佳工艺:液料比为1∶20,提取时间为3 h,提取温度为100℃,提取次数为3次.长白山野生牛肝菌多糖对羟基自由基的清除率(68.37%~96.82%)在200~800μg/m L剂量下强于水溶性维生素E(65.51%~83.21%).结论确定了长白山野生牛肝菌多糖提取的最佳工艺条件,并检测其体外抗氧化能力,为进一步探究长白山野生牛肝菌多糖的药理作用奠定了基础.
Objective According to L9( 34) orthogonal program,the polysaccharide content was finally used as the index of extraction rate of wild Boletus edulis in Changbai Mountain,detection of its antioxidant activity in vitro.Method The extraction conditions( liquid-to-liquid ratio,extraction time,extraction temperature,extraction times) of wild Boletus edulis fruit bodies in Changbai Mountain were optimized to detect their antioxidant activities in vitro. The wild Boletus edulis in Changbai Mountain was used as the raw material. The ability of extracting polysaccharides from wild Boletus edulis in Changbai Mountain by using water-soluble VE and distilled water as solvent in distilled water as solvent was determined by salicylic acid method.Results The effects of four factors on the extraction rate of wild Boletus edulis polysaccharides in Changbai Mountain were as follows: A,R =2.66>extraction time( B,R = 1.73) >extraction temperature( C,R = 1.02) >extraction times( D,R = 0.19),the optimal process for extracting polysaccharides from wild Boletus edulis in Changbai Mountain was determined as liquid to material ratio 1 ∶ 20,extraction time was 3 h,extraction temperature was 100 ℃,and extraction times were 3 times. The clearance rate of hydroxyl free radicals in wild Boletus edulis polysaccharides in Changbai Mountain( 68.37% ~ 96.82%) was stronger than that of water-soluble VE( 65.51% ~ 83.21%) at the dose of 200~ 800 μg/m L. Conclusion This experiment determined the optimal process conditions for extracting polysaccharides from wild Boletus edulis in Changbai Mountain,and tested its antioxidant capacity in vitro,which can lay a foundation for further exploration of the pharmacological effects of wild Boletus edulis polysaccharides in Changbai Mountain.
