二维逆流色谱技术研究进展 被引量:2

Two-dimensional counter current chromatography——advances of recent technologies and key issues

摘要 一维色谱因其有限的峰容量和相对较低的分离效率而无法一次将目标组分从复杂样品中分离出来。二维色谱因具有峰容量高,分辨率强,进样量大以及选择性强等特点而在天然产物、蛋白质等多组分分离中得到广泛应用。二维逆流色谱(2DCCC)研究目前主要包括在线或离线模式下逆流色谱-逆流色谱联用(CCC-CCC)、逆流色谱-液相色谱联用(CCC-LC),聚焦于2个维度色谱柱的快速选择、溶剂系统的最优条件确定以及与其他技术的联合使用等问题。相比于传统二维LC,CCC与LC的联用在流动相溶剂系统兼容性、正交性程度、纯化能力方面均显示出更大的优势。本文综述了近年来2DCCC技术最新应用研究进展,对关键问题和理论进行了分析,并对2DCCC技术的发展趋势进行展望。 One dilemma of one-dimensional chromatography is its restriction in separating target components from the complex samples within one time due to its limited peak capacity and low resolution.The two-dimensional counter current chromatography(2 DCCC)owns the merits of high peak capacity,high resolution,large loading capacity and large range of selectivity and has been widely applied in isolating multiple components from natural products,proteins or other complex samples.Two-dimensional counter current chromatography(2 DCCC)studies currently include countercurrent chromatography-countercurrent chromatography(CCC-CCC)and countercurrent chromatography-liquid chromatography(CCC-LC)in on-line or off-line mode,focusing on two dimensional columns,the determination of optimal conditions for solvent systems,and coupling with other technologies.Compared with traditional two-dimensional LC,the combination of CCC and LC shows greater advantages in mobile phase solvent system compatibility,degree of orthogonality,and purification ability.This article reviews the recent application research progress of 2 DCCC technology,analyzes the key issues and theories,and forecasts the development trend of 2 DCCC technology.
