晶种生长法制备生物模拟酶金纳米棒

摘要 分别制备金纳米棒种子溶液和生长溶液,然后采用晶种生长法制备了可调控的生物模拟酶金纳米棒,探讨了NaBH 4用量、搅拌方式、反应温度对种子溶液的影响以及油酸钠(NaOL)对生长溶液的影响,通过TEM、UV Vis等对金纳米棒的形貌、性质进行了表征。结果表明,在27℃、700 r·min^-1搅拌条件下,将0.5 mL 0.5 mmol·L^-1 HAuCl 4溶液与0.5 mL 0.2 mol·L^-1 CTAB溶液(50℃温水溶解)混合于烧杯中,得到Au(Ⅲ)CTAB溶液;然后将0.06 mL 0.01 mol·L^-1新鲜的NaBH 4溶液用水稀释到0.1 mL,注入到Au(Ⅲ)CTAB溶液中,完全混合后,再在1200 r·min^-1搅拌2 min,得棕黄色种子溶液。将5 mL 0.2 mol·L^-1 CTAB溶液与0.15 mL 4 mmol·L^-1 AgNO 3溶液混合于烧杯中,加入5 mL 1 mmol·L^-1 HAuCl 4溶液,30℃静置15 min,溶液呈亮黄色,再缓慢搅拌90 min,加入0.07 mL 78.8 mmol·L^-1 AA溶液,1200 r·min^-1剧烈搅拌30 s,黄色消失,得无色透明生长溶液。向生长溶液中加入0.012 mL种子溶液,30℃静置12 h,得到纵向等离子共振吸收峰为550~800 nm,尺寸(长×宽)为(37 nm×12 nm)^(48 nm×16 nm)粒径均一的酒红色金纳米棒。 After the preparation of gold nanorods(GNRs) seed solution and growth solution,we obtained the controlled biomimetic enzyme GNRs by a seed growth method.Moreover,we investigated the effects of NaBH4 amount,stirring method,reaction temperature on seed solution and the effect of NaOL on growth solution,and characterized the morphology and performance of GNRs by TEM and UV-Vis.The optimum preparation conditions of seed solution are determined as follows:Au(Ⅲ)-CTAB solution is obtained by mixing 0.5 mL 0.5 mmol·L^-1 HAuCl4 solution with 0.5 mL 0.2 mol·L^-1 CTAB solution(50 ℃ warm water dissolved) in a beaker at 27 ℃ and stirring at a rate of 700 r·min-1;the brown-yellow seed solution is obtained by diluting 0.06 mL 0.01 mol·L^-1 fresh NaBH4 with water to 0.1 mL,then injecting the dilution into the Au(Ⅲ)-CTAB solution,and stirring for 2 min at 1 200 r·min^-1.The optimum preparation conditions of growth solution are determined as follows:mixing 5 mL 0.2 mol·L^-1 CTAB solution with 0.15 mL 4 mmol·L^-1 AgNO3 solution in a beaker,adding 5 mL 1 mmol·L^-1 HAuCl4 solution,standing for 15 min at 30 ℃,slowly stirring for 90 min,adding 0.07 mL 78.8 mmol·L^-1 AA solution,vigorously stirring for 30 s at 1 200 r·min^-1,the yellow disappears and the colorless transparent growth solution is obtained.Finally,after adding 0.012 mL seed solution into growth solution,and standing for 12 h at 30 ℃,the uniform wine red GNRs is obtained with vertical plasma resonance absorption peak of 550 nm to 800 nm and particle size(length×width) of 37 nm×12 nm to 48 nm×16 nm.
