响应面法优化大孔树脂纯化黄精总皂苷提取物工艺 被引量:12

Process of Macroporous Resin for Purification of Total Saponins in Polygonatum sibiricum Extract by Response Surface Methodology

摘要 为优化大孔树脂纯化黄精皂苷提取物的工艺参数,以黄精皂苷提取物为原料,考察6种大孔树脂对黄精皂苷提取物中皂苷的静态吸附与洗脱性能后,选择采用AB-8型大孔树脂纯化提取物中总皂苷。利用树脂动态吸附-洗脱,通过响应面回归方程预测最佳纯化工艺为:体积60 mL的0.57 mg/mL黄精皂苷提取物,以2.0 mL/min流速上样至AB-8型大孔树脂内吸附后,采用体积90 mL的80.81%乙醇溶液,以1.62 mL/min流速洗脱。该纯化工艺的树脂泄漏与洗脱曲线表明流出液总皂苷浓度低于树脂泄漏点,且洗脱物的峰单一、对称并尖锐,利用紫外-可见分光光度法定量分析显示,提取物中总皂苷含量由纯化前2.75 mg/g提高至纯化后7.45 mg/g,因此该纯化工艺方便可行,适于相关行业推广。 To optimize the technical condition for purification of total saponins of Polygonatum sibiricum with macroporous resin,the properties of static adsorption and elution of six kinds of macroporous resins were investigated.The result showed that AB-8 macroporous resin was confirmed as the experimental resin.Using dynamic adsorption and desorption experiments,the optimum purification condition was obtained as follows:the sixty milliliter of 60 mL 0.57 mg/mL Polygonatum sibiricum extract was loaded on to the resin column at flow rate of 2.0 mL/min and eluted with ninety milliliter of 80.81%ethanol at flow rate of 1.62 mL/min by regression equation of response surface methodology.Under this condition,the concentration of total saponins of effluent in loading sample was less than resin leaking point from leaking curve observation and quantitative elution of single material as one sharp and symmetry peak.The content of total saponins of Polygonatum sibiricum extract was increased from 2.75 mg/g to 7.45 mg/g by quantitative analysis of UV-visible spectrometry.Optimization of the purification process by RSM of total saponins in Polygonatum sibiricum was convenient and feasible.
