摘要
目的探讨组蛋白去乙酰化酶抑制剂(HDACi)SAHA对EL4细胞中miR-150表达及增殖的影响。方法使用HDACi SAHA处理EL4细胞,定量RT-PCR法测定各组细胞中miR-150的表达水平,MTT法测定各组细胞增殖能力。结果经HDACi SAHA处理后, EL4细胞中miR-150的表达随着SAHA剂量的增加而增加。经HDACi SAHA处理后,EL4细胞增殖能力下降。结论 HDACi SAHA能够恢复或提高小鼠T淋巴瘤细胞EL4中miR-150表达并抑制EL4细胞增殖。miR-150在EL4细胞中表达可能受乙酰化修饰的调控,乙酰化修饰调控亦可能通过miR-150下调EL4细胞的增殖能力。
Objective To investigate the effect of histone deacetylase inhibitor(HDACi) SAHA on expression of miR-150 and proliferation of EL4 cells. Methods Quantitative RT-PCR was used to detect the expression of microRNA-150 in EL4 cells treated with HDACi SAHA. Cell proliferation was measured by MTT. Results The expression of miR-150 increased with the increase of SAHA dose. After treatment with SAHA, the proliferation ability of EL4 cells decreased. Conclusion Histone deacetylase inhibitor SAHA can increase the expression of microRNA-150 in EL4 cells and inhibit the cell proliferation, which may be associated with the down-regulation of acetylation modification by miR-150.
