盐酸舍曲林有关物质的合成

摘要 目的:为控制盐酸舍曲林的质量,合成原料中3种有关物质.方法:以1-萘酚为起始原料经过Friedel-Crafts反应、Leuckart反应后成盐得到了有关物质B和有关物质C.又3-氯苯乙酮为起始原料经过Aldol反应、还原、Friedel-Crafts反应、Leuckart反应后成盐得到了有关物质D.结果和结论:合成的盐酸舍曲林原料中3种有关物质,并经过1H-NMR和ESI-MS等确证结构,纯度经HPLC检测均在99%以上,这些有关物质的合成可作为盐酸舍曲林质量控制中的杂质对照品,从而对杂质进行准确地定量、定性分析. Objective: To perform the quality control of Sertraline hydrochloride, three related substances were synthesized. Methods: The realated substance (B) and (C) were synehesized by the Friedel-Crafts reaction, Leuckart reaction started from compound 1- Naphthol. The realated substance (D) was synehesized by the Aldol reaction, reduction,Friedel-Crafts reaction、Leuckart reaction and salification, started from compound 3-Chloroacetophenone. Results and Conclusion: Three related substances were synthesized and their chemical structure were confirmed by 1H-NMR and MS.3Theirpurities wree over 99% by the detection of HPLC, and they can be used as the reference substances in the quality control of sertraline hydrochloride.
