宣肺平喘方对哮喘大鼠肺、肠组织病理形态变化及Th1/Th2影响的研究 被引量:3

摘要 目的:观察宣肺平喘方对哮喘大鼠肺、肠组织病理形态及Th1/Th2平衡的影响,为中医“肺与大肠相表里”理论提供支持。方法:将40只SD大鼠,随机均分为正常组、模型组、地塞米松组、宣肺平喘方组。通过腹腔注射OVA与氢氧化铝致敏、OVA雾化吸入激发制备哮喘大鼠模型。给药2周,采用HE染色观察大鼠肺、肠病理变化,Elisa法检测Th1、Th2相关细胞因子IL-4、IL-5、IFN-γ、IL-2含量。结果:正常组肺肠组织均未见炎性病理改变;模型组肺部可见典型支气管哮喘病理表现,肠组织可见炎细胞浸润,肌层增厚;宣肺平喘方组病理变化明显减轻,与地塞米松组相近。模型组大鼠血Th1相关因子IFN-γ、IL-2含量明显下降,而宣肺平喘方组表达上升;模型组大鼠血Th2相关因子IL-4、IL-5含量明显上升;而宣肺平喘方组表达下降,差异具有统计学意义(P<0.05)。结论:宣肺平喘方能减轻哮喘大鼠肺、肠组织病理损伤,为中医“肺与大肠相表里”理论提供实验基础;并通过改变Th1、Th2相关细胞因子IL-4、IL-5、IFN-γ、IL-2含量调节Th1/Th2平衡,纠正免疫功能紊乱。 Objective:To observe the effects of Xuanfei Pingxhuan Fang on pathological morphology of lung and intestine in asthmatic rats and Th1/Th2 so as to provide support for the theory of“lung and large intestine superficial and interior”.Methods:40 SD rats were randomly divided into four group:normal group,model group,Dexamethasone group and Xuanfei Pingchuan Fang group.Asthmatic rats were induced by intraperitoneal injection of OVA,sensitized with aluminum hydroxide and inhaled by OVA.The pathological changes of lung and intestine were evaluated by HE staining.Detection of Th1,Th2-related cytokines IL-4,IL-5,IFN-γand IL-2 by Elisa.Results:No inflammatory pathological changes were found in the lung and intestine in the normal group,inflammatory cell infiltration and myometrial thickening were observed in the intestinal tissue in model group that showed pathological changes of typical bronchial asthma,inflammatory changes were relieved obviously in Xuanfei Pingxhuan Fang group,that was almost the same as Dexamethasone group.The levels of serum Th1-related factors IFN-γand IL-2 in model group decreased significantly,while the expressions of Xuanfei Pingchuan Fang group increased;the levels of serum Th2-related factors IL-4 and IL-5 in model group increased significantly;while the expressions of Xuanfei Pingchuan Fang group decreased,with statistical significance(P<0.05).Conclusion:Xuanfei Pingxhuan Fang can alleviate the pathological damage of lung and intestine in asthmatic rats,provide experimental basis for the theory of“lung and large intestine superficial and internal”,and Th1/Th2 balance was regulated by altering the contents of Th1,Th2-related cytokines IL-4,IL-5,IFN-γand IL-2 to correct immune dysfunction.
