摘要
为了提高猪圆环病毒2型(PCV2)抗原的病毒含量,对PCV2片状载体悬浮培养生产工艺进行了试验。将PCV2接种到PK-15细胞悬液中,在转瓶上培养,第2次带毒传代后,接种到片状载体上悬浮培养,当培养液中耗糖量稳定时,换维持液,每48 h收获1次,可以连续收获5次以上,有效抗原含量比传统转瓶生产工艺提高了5~10倍。
In order to improve the virus content of porcine circovirus type 2(PCV2)antigen,the production process of PCV2 flake carrier suspension culture was studied.PCV2 was inoculated in a PK-15 cell suspension and cultured on roller bottles.After the second passage,it was inoculated on a flake carrier for suspension culture.When the sugar consumption in the culture solution was stable,the cell maintenance solution was replaced.Once harvested every 48 hours,it could be harvested more than five times in a row,and the effective antigen content was 5-10 times higher than the traditional roller bottle production process.
