老年皮肤恶性黑色素瘤病人基质金属蛋白酶、内皮生长因子及相关血清因子水平分析

Analysis of matrix metalloprotease,endothelial growth factor and related serum factors in elderly patients with cutaneous malignant melanoma

目的探讨老年皮肤恶性黑色素瘤病人基质金属蛋白酶(MMPs)、内皮生长因子(VEGF)及相关血清因子的表达水平。方法选取我院收治的35例老年恶性黑色素瘤病人作为黑色素瘤组,同期的35例老年普通色素痣病人作为对照组。采用免疫组化技术检测MMP-2、MMP-9、金属蛋白酶组织抑制剂-2(TIMP-2)、VEGF-A、VEGF-C、血管内皮生长因子受体-3(VEGFR-3)在2组病人中的阳性表达率,酶联免疫吸附法测定白细胞介素-23(IL-23)及S100蛋白含量。结果黑色素瘤组MMP-2、MMP-9、TIMP-2、VEGF-A、VEGF-C及VEGFR-3的阳性表达率与对照组相比,均显著升高(P<0.05);IL-23及S100蛋白含量也显著高于对照组(P<0.05),且均随着分期升高而升高。结论老年皮肤恶性黑色素瘤病人存在MMPs及其抑制剂、VEGF及其受体、IL-23及S100蛋白的表达异常,临床中应加强对此类因子的检测,以便准确判断病人病情。

Objective To explore the levels of matrix metalloproteinases(MMPs),endothelial growth factors(VEGF)and related serum factors in the elderly patients with cutaneous malignant melanoma.Methods Thirty-five elderly patients with malignant melanoma in our hospital were selected as the melanoma group,while 35 elderly patients with common pigmented nevus were selected as the control group.Immunohistochemical staining was used to detect the positive expression rates of MMP-2,MMP-9,tissue inhibitors of metalloproteinase-2(TIMP-2),VEGF-A,VEGF-C and VEGFR-3 in the two groups.IL-23 and S100 protein content was detected by ELISA method.Results The positive expression rates of MMP-2,MMP-9,TIMP-2,VEGF-A,VEGF-C and VEGFR-3 in the melanoma group were significantly higher than those in the control group(P<0.05).And the levels of IL-23 and S100 protein were also significantly higher than those in the control group(P<0.05),and all of them increased with the increase of stage.Conclusions The expression of MMPs and its inhibitors(VEGF)and its receptors(IL-23 and S100 protein)are abnormal in the elderly patients with malignant melanoma.The detection of such factors should be strengthened in clinical practice.
