摘要
目的观察急性百草枯中毒大鼠肺组织中肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)、白细胞介素-13(IL-13)表达情况,探讨百草枯中毒的机制及血必净注射液的干预作用。方法将84只Wistar大鼠按照随机数字表分为对照组、中毒组和血必净组,每组28只。中毒组和血必净组均腹部注射百草枯染毒,之后对照组和血必净组每天分别注射0.9%NaCl及血必净注射液4 mL/kg,分别于注射后1 d、3 d、7 d、14 d、21 d、28 d、35 d每组各取出4只大鼠,开胸留取肺脏标本,光镜观察肺组织病理结构,免疫组化染色法检测肺组织中TNF-α、TGF-β1、IL-13表达情况。结果中毒组大鼠各时间点肺组织中TNF-α、TGF-β1、IL-13表达水平明显高于对照组(P均<0.05)。血必净组大鼠各时间点肺组织中TNF-α、TGF-β1、IL-13表达水平均明显低于中毒组(P均<0.05),但均明显高于对照组(P均<0.05)。结论TNF-α、TGF-β1、IL-13参与了百草枯中毒肺损伤;血必净注射液能够通过抑制TNF-α、TGF-β1和IL-13的表达改善百草枯中毒导致的肺损伤。
Objective It is to observe the expression of tumor necrosis factor-α(TNF-α),transforming growth factor-β1(TGF-β1)and interleukin-13(IL-13)in lung tissue of rats with acute paraquat poisoning,and to explore the mechanism of paraquat poisoning and the intervention of Xuebijing injection on it.Methods 84 Wistar rats were divided into control group,poisoning group and Xuebijing group according to the random number table,with 28 rats in each group.Both the poisoning group and the Xuebijing group were exposed to paraquat by intraperitoneal injection of paraquat,then the control group and Xuebijing group were injected with 0.9%NaCl and Xuebijing injection 4 mL/kg every day,respectively.In 1 d,3 d,7 d,14 d after injection,4 rats were taken out from each group and their lung specimens were taken from the chest to observe the pathological structure of the lung tissues by light microscopy.Immunohistochemical staining was used to detect the expression of TNF-α,TGF-β1,IL-13 in lung tissue.Results The expression levels of TNF-α,TGF-β1 and IL-13 in the lung tissue of the rats in the poisoning group were significantly higher than those in the control group at every time point(P<0.05).The expression levels of TNF-α,TGF-β1 and IL-13 in lung tissue of Xuebijing group were significantly lower than those in the poisoned group at every time point(P<0.05),but they were all significantly higher than those in the control group(P<0.05).Conclusion TNF-α,TGF-β1 and IL-13 are involved in lung injury induced by paraquat.Xuebijing injection can improve lung injury caused by paraquat poisoning by inhibiting the expression of TNF-α,TGF-β1 and IL-13.
