人热休克蛋白B6在膀胱癌组织中的表达及其预后意义

Expression of Heat Shock Protein B6 and Its Prognostic Role in the Bladder Cancer

目的通过分析癌症基因组数据库(TCGA)中基因芯片数据,挖掘人热休克蛋白B6(HSPB6)在正常膀胱组织及膀胱癌组织中的表达及其预后意义。方法通过Oncomine及人蛋白质数据库分析HSPB6在正常膀胱组织与膀胱癌组织中的差异表达;通过基因表达谱数据动态分析HSPB6表达水平与患者预后的关联;通过String数据库分析与HSPB6相关的基因及蛋白并预测其功能。结果膀胱癌组织中HSPB6的表达较正常膀胱组织低,差异有统计学(P<0.05);生存分析中HSPB6与患者疾病无进展生存时间、总生存时间具有明显相关性(P<0.05)。结论尽管HSPB6在膀胱癌组织中普遍较癌旁正常组织低表达,但其在癌组织中的高表达反而是患者不良预后因素。

Objective To analyze the gene microarray in The Cancer Genome Atlas(TCGA)database,and to investigate the expression and prognostic significance of heat shock protein B6(HSPB6)in the normal bladder tissue and bladder cancer tissue.Methods The expression of HSPB6 in the bladder cancer tissues and the normal bladder tissues were measured by using Oncomine database and Human Protein Atlas.The correlations between HSPB6 expression and prognosis of bladder cancer were analyzed by gene expression profiling interactive analysis.The genes and proteins related to HSPB6 were analyzed and their functions were predicted by String database.Results The expression of HSPB6 in the bladder cancer tissues was lower than that in the normal bladder tissues,and the difference was statistically significant(P<0.05).In survival analysis,HSPB6 was significantly correlated with disease free survival time and overall survival time of patients(P<0.05).Conclusion Although HSPB6 is generally lower expressed in the bladder cancer tissues than in the adjacent normal tissues,its high expression in the cancer tissues is a poor prognostic factor of patients.
