摘要
目的探讨胃癌组织中的叉头框蛋白M1(FoxM1)与胃癌患者血清中的α-1-微球蛋白/胰蛋白酶抑制剂前体(AMBP)的联合检测在胃癌诊断及治疗中的临床意义。方法收集105例胃癌患者的血清及肿瘤组织、肿瘤旁组织、正常组织及102名健康体检者的血清,通过免疫组织化学、酶联免疫吸附试验法(ELISA)、蛋白质印迹法等检测胃癌患者与健康体检者血清中AMBP和胃癌患者组织中FoxM1的表达量。结果胃癌患者的AMBP血清平均表达量为(9.14±0.17)μg/ml,高于健康体检者(8.10±0.73)μg/ml(P<0.05)。AMBP血清表达量与患者年龄(P>0.05)、性别(P>0.05)无关,而与肿瘤的分化程度(P<0.05)相关。胃癌组织中FoxM1的表达明显高于癌旁组织(P<0.05)及正常组织(P<0.05)。血清AMBP水平与FoxM1的表达量均与肿瘤的分化程度呈负相关性,肿瘤的分化程度越低,血清AMBP水平与FoxM1的表达量越高。结论AMBP与FoxM1的联合检测为胃癌的诊断及治疗提供新的思路。
Objective To investigate the clinical significance of combined determination of forkhead box M1(FoxM1)in carcinoma tissues and serum alpha-1-microglobulin/bikunin precursor(AMBP)in the diagnosis and treatment of gastric cancer.Methods The serum and carcinoma tissues,adjacent tissues and normal tissues from 105 patients with gastric cancer,and the serum samples from 102 normal subjects were collected.Immunohistochemistry,ELISA and Western blotting were used to determine the serum expression level of AMBP in gastric cancer patients and normal subjects and the tissue expression level of FoxM1 in patients with gastric cancer,thereby providing new ideas for the diagnosis of early gastric cancer.Results The mean expression level of serum AMBP in gastric cancer patients was higher than that in normal subjects[(9.14±0.17)μg/ml vs(8.10±0.73)μg/ml,P<0.05].The serum expression level of AMBP was not correlated with age(P>0.05)and gender(P>0.05),but was correlated with the degree of tumor differentiation(P<0.05).The expression level of FoxM1 in gastric cancer tissues was significantly higher than that in the adjacent tissues(P<0.05)and normal tissues(P<0.05).The serum levels of AMBP and FoxM1 were negatively correlated with tumor differentiation.The lower the degree of tumor differentiation,the higher the expression levels of serum AMBP and FoxM1.Conclusion Combined determination of AMBP and Fox M1 provides new ideas for the diagnosis and treatment of gastric cancer.
