黄芪多糖对糖尿病心肌病模型大鼠的保护作用 被引量:18

Protective effect of Astragalus polysaccharide on diabetic cardiomyopathy model rats

摘要 目的探讨黄芪多糖对糖尿病心肌病模型大鼠的保护作用。方法用高脂饲料联合小剂量链脲佐菌素腹腔注射建立糖尿病心肌病大鼠模型。按随机数表法将大鼠分为5组:对照组、安静组、运动组、药物组和联合组,每组10只。对照组与安静组给予0.9%Na Cl腹腔注射;运动组与联合组给予中等强度跑台训练,21 m·min^-1,坡度2.5°,每日1 h,每周5 d;药物组与联合组给予黄芪注射液30 g·kg^-1腹腔注射,每日1次;连续处理6周,期间实验各组继续以高脂饮食喂养,对照组予以正常饮食。用超声成像系统测定左室射血分数(LVEF),用酶联免疫吸附法测定血清肌酸激酶(CK)水平,用蛋白质免疫印迹法测定心肌组织p38丝裂原活化蛋白激酶(p38MAPK)蛋白表达水平。结果对照组、安静组、运动组、药物组和联合组的LVEF分别为(94.63±4.29)%,(78.72±3.76)%,(82.74±3.17)%,(86.65±2.93)%和(90.02±2.11)%;上述这5组的血清CK水平分别为(321.28±26.74),(520.75±34.18),(483.64±52.49),(427.82±47.15)和(366.48±35.07)U·L^-1;上述这5组心肌组织p38MAPK相对表达量分别为0.49±0.06,1.13±0.15,0.82±0.11,0.71±0.07和0.62±0.06。上述指标:安静组与对照组相比或者运动组、药物组和联合组与安静组相比,差异均有统计学意义(均P<0.05)。结论黄芪多糖能明显改善糖尿病心肌病模型大鼠的心功能,减轻心肌损伤,其作用可能与抑制MAPK信号通路活性有关。 Objective To analysis the protective effect of Astragalus polysaccharide injection(API)on diabetic cardiomyopathy model rats.Methods The diabetic cardiomyopathy model was established by high-fat feed and intraperitoneal injection of low dose streptozotocin.The rats were randomly divided into five groups:control group,rest group,exercise group,drug group and combination group,each group with 10 rats.The control group and rest group were gi-ven intraperitoneal injection of physiological saline,the exercise group and combination group were given moderate intensity treadmill training,21 m·min^-1,slope 2.5°,1 h a day,5 d a week;the drug group and combination group were given intraperitoneal injection of API 30 g·kg^-1,once a day;the treatment lasted for 6 weeks,during which the experimental groups continued to be fed on a high-fat diet,while the control group was given a normal diet.The left ventricular ejection fraction(LVEF)were detected by ultrasound imaging system.The serum creatine kinase(CK)levels were detected by enzyme-linked immunosorbent assay.The p38 mitogen-activated protein kinase(p38MAPK)protein expression levels in myocardium were detected by Western blot.Results The LVEF in control group,rest group,exercise group,drug group and combination group were respectively(94.63±4.29)%,(78.72±3.76)%,(82.74±3.17)%,(86.65±2.93)%and(90.02±2.11)%;the serum CK levels in the above5 groups were respectively(321.28±26.74),(520.75±34.18),(483.64±52.49),(427.82±47.15)and(366.48±35.07)U·L^-1;the p38MAPK protein relative expression levels in the above 5 groups were respectively 0.49±0.06,1.13±0.15,0.82±0.11,0.71±0.07 and 0.62±0.06.Comparison between rest group and control group,or comparison between exercise group,drug group,combination group and rest group,the differences of the indicators were significantly(all P<0.05).Conclusion API can significantly improve cardiac function of diabetic cardiomyopathy model rats,reduce myocardial injury,the effect may be related to the inhibition of the activity of MAPK signaling pathway.
