邻苯二甲酸二甲酯对雄性小鼠生殖系统的影响 被引量:8

Effects of dimethyl phthalate on male mice’s reproductive systems

摘要 目的探讨邻苯二甲酸二甲酯(DMP)的生殖毒性作用。方法以成年雄性ICR小鼠为研究对象,设1000 mg/kg、500 mg/kg、250 mg/kg剂量和对照组连续染毒2个月。测定脏器系数、精子运动及精子数、睾丸细胞酶活力及部分血清生化指标。结果DMP能引起雄鼠500 mg/kg和1 OOOmg/kg染毒组睾丸重量减少(P<0.01),各染毒组附睾重量均减少(P<0.01),500 mg/kg和1000 mg/kg染毒组肝脏肿大(P<0.01);1000 mg/kg染毒组I级、IV级运动精子、精子数与对照组比较差异有统计学意义(P<0.01);各染毒组AKP活力比对照组均显著降低(P<0.01);各染毒组血清Cr、1000 mg/kg染毒组UREA都显著升高,与对照组比较差异有统计学意义(P<0.01)。结论DMP对雄性小鼠具有一定生殖毒性作用,睾丸细胞酶活力的改变,可能与DMP导致睾丸的支持细胞和生精上皮细胞损伤作用有关。 Objective To explore the productive toxicity of the dimethyl phthalate(DMP)in male mice.Methods Adult male ICR mice in different doses(1000 mg/kg,500 mg/kg,250 mg/kg)and the controls were orally administrated with dimethyl phthalate for two months.The viscera coefficients,kinetism of sperm,sperm counts,activity of some enzymes in testis and some serum biochemical index were examined.Results DMP decreased the incidence of mouse testis in the 500 mg/kg group and 1000 mg/kg group(P<0.01).The weight of epididymis decreased in each exposed group(P<0.01).Liver enlargement was observed in the 500 mg/kg and 1000 mg/kg groups(P<0.01).Compared with the control group,the difference in the number of grade I and grade IV motor sperm and total sperm in the 1000 mg/kg group was statistically significant(P<0.01);AKP activity in each group was significantly lower than that in the control group(P<0.01).Serum Cr in each group and UREA in the 1000 mg/kg group significantly increased,and the differences in the control group were statistically significant(P<0.01).Conclusion DMP has some reproductive toxicity on male mice and can influence the activity of testis enzymes,which may be correlated with the impairment of sertoli cell and spermatogenic epithelium cell.
