男性精神分裂症患者吸烟与CYP2C19基因多态性的相关性研究

摘要 目的探讨CYP2C19基因多态性与精神分裂症患者吸烟行为的相关性。方法选取男性精神分裂症患者173例(其中吸烟组86例,非吸烟组87例),男性正常对照组171名(其中吸烟组85名,非吸烟组86名),采用多重SNP分型技术,分型检测CYP2C19基因上的SNP位点rs12248560、rs4986893、rs4244285,分析比较精神分裂症组与正常对照组及吸烟组与非吸烟组在该基因位点的3个SNPs等位基因及基因型频率有无差异。结果*3、*17位点在精神分裂症组与正常对照组之间的基因型分布差异具有统计学意义(P<0.01);*2、*3、*17位点在精神分裂症吸烟组与精神分裂症非吸烟组之间的基因型分布差异具有统计学意义(P<0.01);*1/*2、*2/*2在精神分裂症吸烟组与精神分裂症非吸烟组之间的分布差异具有统计学意义(P<0.05);中间代谢型(IM)、弱代谢型(PM)在精神分裂症吸烟组与精神分裂症非吸烟组之间的分布差异具有统计学意义(P<0.05);*2位点在正常对照吸烟组与正常对照非吸烟组之间的基因型分布差异具有统计学意义(P<0.05)。结论CYP2C19基因多态性与精神分裂症的易感性可能存在关联,且可能与精神分裂症及正常人的吸烟行为具有相关性。 Objective To investigate the relationship between CYP2 C19 gene polymorphism and smoking behavior in patients with schizophrenia.Methods A total of 173 male patients with schizophrenia in schizophrenia group(86 cases in smoking group and 87 cases in non-smoking group),and 171 healthy controls in control group(85 in smoking group and 86 in non-smoking group)were selected.Multiple SNP typing techniques were used,and the SNP loci rs4244285,rs4986893 and rs12248560 on the CYP2 C19 gene were typed and analyzed.3 SNPs alleles and genotypes at the locus between schizophrenia group and control group,as well as between smoking and non-smoking groups,were compared to find whether there existed significant difference in frequency or not.Results There existed significant difference in genotype distributions of *3,*17 locus between schizophrenia group and control group(P<0.01).There existed significant differences in genotype distributions of *2,*3,*17 locus between schizophrenia smoking group and schizophrenia non-smoking group(P<0.01).There existed significant differences in *1/*2,*2/*2 distribution and intermediate metabolite(IM)and poor metabolism(PM)distribution between schizophrenia smoking group and schizophrenia non-smoking group(P<0.05).There existed significant difference in genotype distributions of *2 locus between control smoking group and control non-smoking group(P<0.05).Conclusion The CYP2 C19 gene polymorphism may be associated with susceptibility to schizophrenia and may be associated with smoking behavior in schizophrenia and normal people.
