摘要
目的:通过观察早搏灵对氯化钡诱导的心律失常大鼠的T型钙离子通道α1H亚基表达的影响,评价早搏灵对防治心律失常的作用机制。方法:选用健康大鼠40只,随机分为4组,维拉帕米对照组,生理盐水组,早搏灵胶囊组,模型组,每组10只。记录并比较各组大鼠的心电图变化,采用荧光定量实时PCR技术观察心肌组织T型钙通道基因α1H的变化。结果:早搏灵胶囊组、维拉帕米组,每组心律失常持续时间与模型组相比较均有一定程度的缩短。维拉帕米组T型钙通道α1H亚基为高表达,说明维拉帕米对T型钙通道α1H亚基表达无明显抑制作用;早搏灵胶囊组与模型组相比,T型钙通道α1H亚基为低表达有显著性差异,证明早搏灵胶囊对T型钙通道α1H亚基有抑制作用。结论:早搏灵胶囊能够使氯化钡诱导的心律失常大鼠的T型钙通道α1H亚基表达下调,可缩短心律失常的持续时间,说明早搏灵胶囊治疗心律失常作用机制可能与降低T型钙通道α1H亚基表达相关。
To explore the action mechanism of Zaoboling in preventing and treating arrhythmia by observing its influence toα1 H subunits expression of T-type calcium ion channel in rats with barium chloride-induced arrhythmia.Methods:40 healthy rats were randomly divided into Verapamil group,normal saline group,Zaoboling group and model group,with 10 rats in each group.Electrocardiogram(ECG)changes were observed andα1 H subunits expression of T-type calcium ion channel in myocardial tissue was detected by RT-PCR.Results:The duration of arrhythmia in Zaoboling group and Verapamil group was both decreased compared to that in the model group.α1 H subunits expression of T-type calcium ion channel was increased in Verapamil group and Zaoboling group compared to that in model group;of which there was statistical difference between Verapamil group and Zaoboling group.Conclusion:Zaoboling capsule can increase theα1 H subunits expression of T-type calcium ion channel in rats with barium chloride-induced arrhythmia,and it can reduce the duration of arrhythmia,which shows that the action mechanism of Zaoboling Capsule in treating arrhythmia may be related to increasingα1 H subunits expression of T-type calcium ion channel.
