不同炮制品组方的独活寄生汤对大鼠佐剂性关节炎的作用及炎症因子比较研究

Effect of Duhuo Jisheng Decoction Composed of Different Processed Products on Adjuvant Arthritis in Rats and Its Influence to Inflammatory Factors: A Comparative Study

目的:比较不同炮制品组方的独活寄生汤对大鼠佐剂性关节炎的治疗作用,并探讨其在炎症细胞因子水平上的作用机制。方法:225只大鼠随机分为正常组、模型组、阿司匹林片组、不同炮制品组方的独活寄生汤高、中、低剂量组,完全弗氏佐剂诱导佐剂性关节炎后,测定原发性和继发性足肿胀,观察胸腺及脾脏指数的变化,检测血清炎性细胞因子TNF-α、IL-6的水平和关节液PGE2含量。结果:与模型组相比,第1组中、高剂量组,第2组中、高剂量组,第3组的中、高剂量组,第4组的中剂量组均能抑制大鼠原发性及继发性足肿胀,增加胸腺指数,显著降低血清TNF-α、IL-6水平及关节液PGE2含量,有显著的差异(P<0.05);由盐杜仲、酒炙牛膝、酒川芎、酒当归、酒白芍组方(第四组)的独活寄生汤的高剂量组与模型组比较有极显著的差异(P<0.01)且强于其生品组和其他各组,差异有统计学意义(P<0.05)。结论:独活寄生汤在临床上用于治疗佐剂性关节炎,组方时杜仲用盐制品,牛膝、川芎、当归、白芍用酒炙品,其作用机制可能与增强机体的免疫功能、抑制促炎因子的产生有关。

Objective:To compare the therapeutic effects of Duhuo Jisheng Decoction composed of different processed products on adjuvant arthritis(AA)in rats and explore its mechanism at the level of inflammatory cytokines.Methods:225 rats were randomly divided into the normal group,the model group,Aspirin group and Duhuo Jisheng Decoction groups(low-dose,medium-dose and high-dose)with different processed products.After AA was induced by Freund’s adjuvant,primary and secondary foot swelling were measured,indexes of thymus and spleen were observed,serum levels of inflammatory cytokines,such as TNF-a,IL-6 and PGE2,in synovial fluid were detected.Results:Compared to the model group,the medium-dose and high-dose of group 1,the medium-dose and high-dose of group 2,the medium-dose and high-dose of group 3 and the medium-dose of group 4 could inhibit primary and secondary foot swelling,increase thymus index,significantly reduce serum levels of TNF-a and IL-6,and reduce the content of PGE2 in joint fluid(P<0.05).There was significant differences between the high-dose of group 4(salted Eucommia ulmoides,wined Achyranthes bidentata,wined Ligusticum Chuanxiong and wined Angelica sinensis)and the model group(P<0.01).The high-dose of group 4 was better than the other treatment groups(P<0.01).Conclusion:Duhuo Jisheng Decoction is used to treat AA in clinic.When Eucommia ulmoides is processed with salt Achyranthes bidentata,Ligusticum Chuanxiong and Angelica sinensis are processed with wine can enhance immune function and inhibit the production of proinflammatory factors.
