生槟榔中生物碱与鞣质类成分对斑马鱼急性毒性的比较研究 被引量:2

摘要 目的:通过比较生槟榔中生物碱及鞣质类成分对斑马鱼的急性毒性,初步探讨生槟榔的毒性成分。方法:采用溶剂萃取法和明胶沉淀法分别制备生槟榔的总生物碱、总鞣质部位并对其进行含量测定;采用不同浓度的总生物碱、总鞣质处理发育正常的斑马鱼幼鱼24 h,次日根据斑马鱼死亡情况用Bliss法计算相应的LC50值。结果:溶剂萃取法和明胶沉淀法制得总生物碱、总鞣质的含量分别为6.67 mg/mL和53.74%,且总生物碱中不含鞣质,总鞣质中不含生物碱;总生物碱、总鞣质对斑马鱼幼鱼的LC50值分别136.14μg/mL和21.52μg/mL。结论:生槟榔中总鞣质对斑马鱼幼鱼的急性毒性大于总生物碱,提示鞣质类成分也是生槟榔的毒性成分。 Objective:To explore the poisonous component of raw arecae semen by comparing the acute toxicity of alkaloids and tannins in raw arecae semen to zebrafish.Methods:Alkaloids and tannins of raw arecae semen were prepared by solvent extraction and Gelatin precipitation respectively.Zebrafish were exposed to the aqueous extraction of alkaloids and tannins with different concentrations for 24 h and their LC50 were calculated.Results:The concentration of prepared alkaloids and tannins were respectively 6.67 mg/mL and 53.74%.The LC50 value of alkaloids and tannins to Zebrafish were 136.14μg/mL and 21.52μg/mL.Conclusion:The tannins were the most poisonous component in raw areca semen.
