摘要
为探讨茶花鸡遗传多样性和起源进化,以茶花鸡为试验素材,对30只茶花鸡线粒体DNA D-loop区全序列进行测序,结合GenBank已公布D-loop区全序列的19条红色原鸡序列,进行比对和分析处理。结果显示:茶花鸡线粒体D-loop区全长为1 232 bp,无缺失和插入。30只个体共发现9处单核苷酸多态位点,为3种单倍型。系统发育分析显示,3种单倍型可分为A和B两个分支,A和B分支均与Gallus gallus spadiceus聚为一类,推测这两个分支可能均起源于红色原鸡滇南亚种。研究从分子水平为茶花鸡遗传资源保护和开发利用提供了参考。
To investigate the genetic diversity and origin evolution of Chahua chicken, complete mtDNA D-loop sequences of 30 Chahua chickens were analyzed by PCR amplification and sequencing technique, and compared with the 19 red jungle fowl sequences in GenBank. The results showed that the D-loop region of Chahua chicken was 1 232 bp,without missing and insert. In the 30 samples assayed, 9 mutation sites and 3 haplotypes were found. Phylogenetic analysis showed that 3 haplotypes of Chahua chicken converged into 2 clades. The clade A and clade B were clustered with Gallus gallus spadiceus, indicated that the two clades may originated from the continental subspecies of Gallus gallus spadiceus. The results would be a reference for genetic resources conservation and development and utilization of Chahua chicken.
