摘要
【目的】观察补髓生血颗粒对慢性再生障碍性贫血(CAA)患者的骨髓极迟活化抗原4(VLA-4)和极迟活化抗原5(VLA-5)mRNA表达的影响及其可能的作用机制。【方法】选取30例CAA肾阴阳两虚型患者为试验组,另招募10例无血液疾病及免疫系统疾病的健康志愿者为正常对照组。试验组患者给予补髓生血颗粒治疗,疗程6个月。采用实时荧光定量逆转录-聚合酶链反应(PT-PCR)法检测CAA患者治疗前后骨髓VLA-4和VLA-5 mRNA表达水平,并与正常对照组作比较。【结果】治疗前,CAA患者骨髓VLA-4 mRNA呈低表达状态,VLA-5 mRNA呈高表达状态,与正常对照组比较,差异有统计学意义(P<0.05);应用补髓生血颗粒治疗后,试验组患者VLA-4 mRNA表达水平较治疗前升高,VLA-5 mRNA表达水平较治疗前降低,差异均有统计学意义(P<0.05),但均未恢复到正常对照组水平,差异均有统计学意义(P<0.05)。【结论】CAA患者VLA-4和VLA-5 mRNA的异常表达可能参与CAA的发病过程;补髓生血颗粒可能通过调节VLA-4和VLA-5 mRNA的表达来改善CAA患者骨髓造血功能和恢复造血微环境的状态。
Objective To observe the effect of Busui Shengxue Granules on the mRNA expression of late-activation antigens of very late antigen-4(VLA-4)and very late antigen-5(VLA-5)in the bone marrow of chronic aplastic anemia(CAA)patients,and to explore the possible therapeutic mechanism.Methods Thirty CAA patients with kidney Yin and Yang dual deficiency syndrome served as the trial group,and 10 healthy volunteers without blood diseases or immune system diseases were used as the normal control.The trial group was given oral use of Busui Shengxue Granules for 6 months.The real-time fluorescence quantitative reverse transcription-polymerase chain reation(PT-PCR)was used to detect the VLA-4 and VLA-5 mRNA expression levels in the bone marrow of CAA patients before and after treatment.And the VLA-4 and VLA-5 mRNA expression levels were compared between the two groups.Results Before treatment,VLA-4 mRNA expression level was low and VLA-5 mRNA expression level was high in the bone marrow of CAA patients,the difference being significant compared with those in the normal control group(P<0.05).After treatment with Busui Shengxue Granules,the expression level of VLA-4 mRNA was increased and that of VLA-5 mRNA was decreased in the bone marrow of CAA patients(P<0.05 compared with those before treatment),but their levels did not arrive to the level of the normal control group(P<0.05).Conclusion The abnormal expression of VLA-4 and VLA-5 mRNA in CAA patients may be involved in the pathogenesis of CAA,and Busui Shengxue Granules can regulate the expression of VLA-4 and VLA-5 mRNA,so as to improve the hematopoietic function of bone marrow and restore the state of hematopoietic microenvironment in CAA patients.
