三黄清胃丸治疗幽门螺旋杆菌相关性胃炎主要活性成分及潜在靶点的网络药理学分析

Analysis of Main Active Components and Potential Targets of Sanhuang Qingwei Pills in Treating Helicobacter Pylori-Associated Gastritis Based on Network Pharmacology

【目的】运用网络药理学研究方法对三黄清胃丸治疗幽门螺旋杆菌相关性胃炎(HAG)主要活性成分及核心靶点进行预测分析。【方法】采用中药系统药理学分析平台(TCMSP)、反向分子对接服务器(DRAR-CPI)和Unipro数据库筛选出三黄清胃丸组成药物的有效活性成分及作用靶点基因,通过GeneCards数据库收集HAG疾病基因,然后对药物作用基因及疾病相关基因进行韦恩分析,确定药物作用潜在靶点,进而构建活性成分-靶点相互作用网络,并进行基因本体论(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析。【结果】研究得到140个作用于HAG疾病靶点的活性成分和94个作用靶点,GO功能富集分析确定了350个条目,KEGG通路分析共发现89条作用通路。【结论】运用网络药理学研究方法预测出三黄清胃丸治疗HAG的主要活性成分及核心靶点。

Objective To predict and analyze the main active components and core targets of Sanhuang Qingwei Pills in treating Helicobacter pylori-associated gastritis(HAG)by the method of network pharmacology.Methods The effective active components and action target genes of the constituted drugs from Sanhuang Qingwei Pills were screened out from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),adverse reactions via the chemical-protein interactome(DRAR-CPI)molecular docking platform and Uniprot database.HAG-related genes were collected from Gene Cards database.And then the drug-related genes and disease-related genes were given Venn analysis to identify potential targets of drug action to construct the component-target interaction networks,and to carry out gene ontology(GO)function enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis.Results One hundred and forty active components associated with HAG targets and 94 action targets were achieved,350 items were identified by GO functional enrichment analysis,and 89 pathways were identified by KEGG pathway analysis.Conclusion The main active components and core targets of Sanhuang Qingwei Pills in treating HAG are predicted and analyzed by the network pharmacology.
